Abstract

Biosynthesis of thyroid hormones is an oxidative process that generates reactive oxygen species and involves thyroperoxidase (TPO). At present a linkage between oxidative stress and triggering of autoimmune reactions has been reported, which emphasizes the importance of increased reactive oxygen species generation in condition of thyroid peroxidase hyperactivity. To verify this hypothesis, a relationship between TPO enzymatic activity and its avidity in patients with autoimmune thyroiditis was determined. The linkage between diagnosis and the enzyme parameters shows that this alterations in TPO function may lead to reduction of oxidation disbalance which in turn may change TPO accessibility for an immune system.

Активные формы кислорода участвуют в биосинтезе тиреоидных гормонов. В нашей лаборатории установлена связь между активностью тиреопероксидазы (ТПО) и авидностью (косвенная оценка количества антигенных детерминант на миллиграмм белка) у пациентов с аутоиммунными тиреоиолигами. Обнаруженная нами корреляция диагноза с исследуемыми параметрами фермента каждого конкретного пациента подтверждает гипотезу, согласно которой модификация специфической активности и авидности может характеризовать изменение конформации ТПО, приводящее к нарушению окислительно-восстановительного равновесия иодирующей системы, повреждению апекальной мембраны тиреоцита, что может быть причиной доступности фермента для иммунной системы.
