Abstract

Aplicaciones biotecnológicas como la Reacción en Cadena de la Polimerasa Cuantitativa (RT-qPCR) son técnicas de laboratorio que han revolucionado el mundo de la Biología Molecular, proporcionando resultados en menor tiempo y un alto grado de confiabilidad, permitiendo la comprensión de los genes, funcionamiento y expresión, como es el caso de los genes de las antocianinas de las cuales en los últimos tiempos se ha incrementado su interés debido a su actividad antioxidante, la cual genera efectos terapéuticos como reducción de enfermedades coronarias, mejoramiento de la agudeza visual, efectos anti-cancerígenos, anti-envejecimiento y antitumorales. Mediante la Reacción en Cadena de la Polimerasa con transcripción inversa (RT-qPCR), se logró analizar el nivel de expresión de los genes β-Actin, RuANS y RuMYB10, involucrados en la biosíntesis de antocianinas en Rubus niveus, recolectadas en tres sectores del Cantón Rumiñahui. De acuerdo a los valores obtenidos con los análisis estadísticos Shapiro Wilks, Anova y el test de Tukey, se observó que no existen diferencias significativas en la expresión de los tres genes, generando los siguientes resultados: gen β-Actin presenta mayor concentración (174,65ng/mL), seguido por el gen RuANS (167,43ng/mL) y finalmente el gen RuMYB10 (163,55ng/mL), llegando a la conclusión que la especie estudiada presenta un nivel de expresión similar entre los tres genes analizados. La presente investigación puede ser considerada como punto de partida para proyectos de mejoramiento de las especies del género Rubus (mora), con el propósito de incrementar su valor nutritivo y cultivarlas como fuente para la producción de nutracéuticos, medicamentos y por ende desarrollar la productividad y valor económico que este cultivo representa para el país.

Biotechnological applications such as (RT-qPCR) Quantitative Polymerase Chain Reaction, are laboratory techniques that have revolutionized the world of Molecular Biology, providing results in less time and in a high degree of reliability, allowing the understanding of genes, functioning and expression, as in the case of the genes of the anthocyanins, which in recent times have increased their interest due to its antioxidant activity which generates therapeutic effects such as reduction of coronary heart disease, improvement of visual acuity, anti-cancer, anti-aging and antitumor effects. The Polymerase Chain Reaction with reverse transcription (RT-qPCR) allowed to analyze the level of expression of β-Actin, RuANS and RuMYB10, genes involved in the biosynthesis of anthocyanin in Rubus niveus, collected in three different sectors of Rumiñahui. According to values obtained with statistical analyses such as Shapiro Wilks, Anova and Tukey test, it was noted that significant differences in the expression of three genes did not exist and there were generated the following results: gene β-Actin, which has greater concentration (174, 65ngmL), followed by the RuANS gene (167, 43ngmL) and finally the RuMYB10 gene (163, 55ngmL), reaching the conclusion that the studied species presents a level of similar expression among the three analyzed genes. This research can be considered as a starting point of projects for improving the genus Rubus (blackberry), in order to increase their nutritional value moreover grow them as a source for the production of nutraceuticals, drugs and therefore developing productivity and economic value that this crop represents for the country.
