The effects of hypoxia, insulin and insulin-like growth factor 1 (IGF-1) on the Ruk/CIN85 expression are studied. It is demonstrated that the Ruk/CIN85 expression in human breast adenocarcinoma MCF-7 cells is not changed under mild hypoxia but induced upon insulin and IGF-1 treatment. Moreover, the upregulation of Ruk/CIN85 increased the MCF-7 sensitivity to IGF-induced proliferation. Neither induction of Ruk/CIN85 expression nor increase in MCF-7 proliferative activity was observed in the cells pretreated with PI3K/Akt pathway inhibitor LY294002. Therefore, our study revealed new factors that influence Ruk/CIN85 expression and biological activity.
