Zelena analitička kemija u studijama stabilnosti etrasimoda i ritlecitiniba

Abstract

Etrasimod i ritlecitinib su novi lijekovi za liječenje upalnih i autoimunih bolesti. Etrasimod, selektivni modulator receptora sfingozin-1-fosfata, koristi se za ulcerozni kolitis, dok je ritlecitinib, inhibitor Janus kinaze 3, učinkovit u liječenju alopecije areate i Crohnove bolesti. S obzirom na sve veći naglasak na održivosti u farmaceutskoj industriji, načela zelene analitičke kemije (ZAK) integrirana su u razvoj analitičkih metoda. Obje studije koristile su UHPLC-DAD-MS/MS tehnike uz validaciju prema ICH smjernicama, uz primjenu ZAK alata poput Analytical Eco-Scale, AMGS-a, ComplexGAPI-ja i AGREE-a. Metoda za etrasimod postigla je visoku preciznost i osjetljivost uz minimalnu upotrebu štetnih otapala. Na Analytical Eco-Scale-u ocijenjena je kao „izvrsna“ (više od 75 bodova), dok je AGREE alat dodijelio visoku ocjenu (0,88/1). Optimizacija mobilne faze i minimalizacija otpada pridonijeli su ekološkom profilu metode. Za ritlecitinib je korištena metrika plave kemije uz zelenu, koja obuhvaća potrošnju vode, energije i upravljanje otpadom. Iako je metoda ocijenjena kao „prihvatljiva“ na Eco-Scale-u (50–75 bodova) i dobila solidnu ocjenu na AGREE-u (0,80/1), pokazala je veću potrošnju energije i otapala u usporedbi s etrasimodom. Metoda za određivanje etrasimoda pokazala je bolje rezultate u ocjeni zelenih metrika zahvaljujući optimiziranim uvjetima kromatografije i manjoj potrošnji resursa. Obje metode naglašavaju smanjenje opasnog otpada i korištenje ekološki prihvatljivih reagensa. Primjena ZAK-a u analizi stabilnosti etrasimoda i ritlecitiniba naglašava važnost održivosti u farmaceutskoj industriji. Metoda za etrasimod pokazala je bolji ekološki profil, dok rezultati za ritlecitinib ukazuju na potrebu za daljnjom optimizacijom.
