Optimizacija kvantitativnog određivanja fungicida video-denzitometrijom

Abstract

Promatran je utjecaj promjene parametara video-denzitometrijskog sistema na kvalitetu slike. Izvedba CAMAG Video Documentation System mjeri gašenje fluorescencije za modelni spoj (fungicid-atrazin), u području UV spektra.Promatran je utjecaj različitog namještanja kamere (promjena otvora zaslona i broja okvira) na odziv kromatografske podloge, baznu liniju šuma, osjetljivost i ponovljivost. Pri radu su korištene standardne pločice (TLC) i pločice visoke djelotvornosti (HPTLC). Rezultati su pokazali da povećanjem otvora zaslona i broja okvira, znači povećanjem svjetline slike, raste odziv podloge i osjetljivost. Osjetljivost postiže maksimum pri umjerenom osvjetljenju pločice, dok daljnje povećanje okvira uzrokuje smanjenje osjetljivosti. Sličan model je promatran mjerenjem granice detekcije (LOD). Ako je F-broj (otvor zaslona) prevelik slika je presvijetla, ako je broj otvora premali slika je pretamna. Oba ekstrema uzrokuje visoki LOD. Ipak, dobra osjetljivost ne znači uvijek visoku granicu detekcije, zbog velikog šuma u istom području mjerenja. ponovljivost snimanja bolja je na tamnije ili umjereno osvjetljenoj pločici. Na HPTLC silikagelnoj podlozi ponovljivost određivanja u uvjetima umjerenog ili slabog osvjetljenja pločice je bolja i kreće se od 1, 5-2, 0 %, a uz veće osvjetljenje raste i do 8 %.
