A RELAÇÃO DOS MIRNA-15-A E 16-1 NA LEUCEMIA LINFOCÍTICA CRÔNICA, E A UTILIZAÇÃO DA ENGENHARIA BIOMÉDICA NO DIAGNÓSTICO: UMA ABORDAGEM DA MEDICINA TRANSLACIONAL / THE RELATIONSHIP OF MIRNA-15-A AND 16-1 IN CHRONIC LYMPHOCYTIC LEUKEMIA, AND THE USE OF BIOMEDICAL ENGINEERING IN DIAGNOSIS: A TRANSLATIONAL MEDICINE APPROACH

Abstract

A leucemia linfocitica cronica (LLC) o tipo mais comum entre adultos no mundo ocidental, surge de um clone maligno de celulas B e pouco se sabe a respeito do seu inicio e progressao. Varios fatores que podem predizer o curso clinico foram identificados, onde as celulas leucemicas tem pouca ou nenhuma mutacao no gene da regiao variavel da cadeia pesada de imunoglobulina (IgVH). A delecao que mais ocorre no DNA genomico em LLC e no cromossomo 13q13, esta delecao e evidente em cerca de 50 por cento dos casos e esta associada ao longo intervalo entre o diagnostico e a necessidade de tratamento. A delecao 13q13 e geralmente a unica anormalidade em LLC e alguns tipos de câncer, evidenciando patogenicidade para o gene excluido. Os miRNAs sao pequenas moleculas de RNAs nao codificantes, com cerca de 19 a 25 nucleotideos que atuam na regulacao genica e estao intimamente ligados a diversas patologias como a diabetes, doencas neurodegenerativas e o câncer. Os miRNAs-15-a e 16-1, encontrados na menor regiao da exclusao em 13q13, sao frequentemente excluidos ou regulados negativamente na LLC. A perda destes miRNAs resulta no aumento de expressao do gene anti-apoptotico Bcl-2. O gene BCL2 anti-apoptotico e hiperexpresso em celulas B da LLC, com isso e possivel identificar que e alvo dos miRNAs 15-a e miR-16-1 e que sua regulacao negativa desencadeia a apoptose. Este estudo teve como objetivo analisar a relacao dos miRNAs 15-a e 16-1 com a LLC. A pesquisa da literatura foi realizada considerando os preceitos de utilizacao dos materiais para o desenvolvimento da pesquisa; embasada em artigos de revisao sistematica, relatos de casos e artigos originais, utilizou-se como base de dados, Medline, Scielo, PubMed, Annual Reviews, Elsevier. De acordo com a analise realisada neste estudo, podemos concluir que os miRNAs 15-a e 16-1, pro-apoptoticos, encontram-se hipoexpressos na Leucemia Linfocitica Cronica.
