The Association of pre-miR27a Gene Polymorphism and Clinicopathological Data in Thai Breast Cancer Patients

Abstract

MiR27a joue un rôle important dans la carcinogenèse, la prolifération cellulaire, l'apoptose, l'invasion, la migration et l'angiogenèse. Plusieurs études ont identifié un rôle important du polymorphisme pré-miR27a (rs895819) A>G dans plusieurs types de cancer. Cette recherche vise à étudier l'association de la prédisposition au cancer du sein pré-miR27a (rs895819) A>G et du cancer du sein, les données clinico-pathologiques et la survie. Des échantillons d'ADN sanguin de 143 patientes thaïlandaises atteintes d'un cancer du sein et de 100 femmes thaïlandaises en bonne santé ont été étudiés pour le polymorphisme pré-miR27a (rs895819) A>G en utilisant le polymorphisme de longueur de fragment de restriction par réaction en chaîne de la polymérase (PCR-RFLP). Les résultats ont révélé que la fréquence des génotypes pré-miR27a (rs895819) A>G n'était pas statistiquement significative entre les patientes atteintes d'un cancer du sein et les sujets témoins normaux. Le génotype rs895819 A>G était significativement associé au paramètre clinicopathologique de différenciation de grade III (P = 0,006), au récepteur de la progestérone (P = 0,011) et au triple négatif (P = 0,031) chez les patientes atteintes d'un cancer du sein, mais pas à la sensibilité au cancer du sein. Le génotype pré-miR27a (rs895819) A>G était significativement associé à un récepteur de la progestérone mal différencié et à un triple négatif chez les patientes atteintes d'un cancer du sein. Par conséquent, pré-miR27a (rs895819) A>G peut être utilisé comme biomarqueur pour un mauvais pronostic.

MiR27a desempeña un papel importante en la carcinogénesis, la proliferación celular, la apoptosis, la invasión, la migración y la angiogénesis. Varios estudios han identificado un papel importante del polimorfismo pre-miR27a (rs895819) A>G en varios tipos de cáncer. Esta investigación tiene como objetivo investigar la asociación de pre-miR27a (rs895819) A>G y la susceptibilidad al cáncer de mama, los datos clinicopatológicos y la supervivencia. Se estudiaron muestras de ADN sanguíneo de 143 pacientes con cáncer de mama tailandés y 100 mujeres tailandesas sanas para determinar el polimorfismo pre-miR27a (rs895819) A>G utilizando el polimorfismo de longitud del fragmento de restricción de la reacción en cadena de la polimerasa (PCR-RFLP). Los resultados revelaron que la frecuencia de los genotipos pre-miR27a (rs895819) A>G no fue estadísticamente significativa entre los pacientes con cáncer de mama y los sujetos de control normales. El genotipo rs895819 A>G se asoció significativamente con el parámetro clínico-patológico de diferenciación de grado III (P = 0.006), receptor de progesterona (P = 0.011) y triple negativo (P = 0.031) en pacientes con cáncer de mama, pero no con susceptibilidad al cáncer de mama. El genotipo pre-miR27a (rs895819) A>G se asoció significativamente con receptor de progesterona poco diferenciado y triple negativo en pacientes con cáncer de mama. Por lo tanto, pre-miR27a (rs895819) A>G se puede utilizar como un biomarcador para el mal pronóstico.

MiR27a plays an important role in carcinogenesis, cell proliferation, apoptosis, invasion, migration and angiogenesis. Several studies have identified an important role of pre-miR27a (rs895819) A>G polymorphism in several types of cancer. This research aims to investigate the association of pre-miR27a (rs895819) A>G and breast cancer susceptibility, clinicopathological data and survival. Blood DNA samples of 143 Thai breast cancer patients and 100 healthy Thai women were studied for pre-miR27a (rs895819) A>G polymorphism using polymerase chain reaction-restriction fragment-length polymorphism (PCR-RFLP).The results revealed that the frequency of pre-miR27a (rs895819) A>G genotypes was not statistically significant different between breast cancer patient and normal control subjects. The rs895819 A>G genotype was significantly associated with clinicopathological parameter of grade III differentiation (P = 0.006), progesterone receptor (P = 0.011) and triple negative (P = 0.031) in breast cancer patients, but not with breast cancer susceptibility.The pre-miR27a (rs895819) A>G genotype was significantly associated with poorly differentiated, progesterone receptor and triple-negative in breast cancer patients. Therefore, pre-miR27a (rs895819) A>G may be used as a biomarker for poor prognosis.
