Purificación y caracterización parcial de la enzima xilanasa a partir del preparado comercial Novoban 240

Abstract

Novoban 240 es un preparado enzimático comercial de la Novo Industri con actividad amilasa, que se obtiene por fermentación sumergida de Bacillus amilolichuefaciens y es utilizado en la industria de elaboración de vinos. Este preparado de uso industrial posee actividad xilanasa, enzima estrechamente relacionada con la respuesta hipersensible de los tejidos vegetales frente al ataque de microorganismos patógenos. En el presente trabajo se establece una metodología de purificación parcial de esta enzima a través del empleo de las cromatografías de intercambio iónico sobre DEAE celulosa y SP Sephadex C-25. El incremento discreto del poder reductor en el medio de reacción durante la hidrólisis enzimática, es un indicador que confirma la presencia de una endoxilanasa en Novoban 240, la que podría ser utilizada para la obtención de xilo-oligosacáridos con actividad inductora de defensa en plantas. El peso molecular estimado de la enzima por electroforesis en gel de poliacrilamida está por debajo de los 14 kDa
