Polimorfizam promotora gena za dopamin-beta-hidroksilazu

Abstract

Patofiziologija posttraumatskog stresnog poremećaja (PTSP) povezuje se s povećanom aktivnošću i osjetljivošću noradrenergičnog sustava. Ključni enzim u sintezi noradrenalina je dopamin-beta hidroksilaza (Dβ H) koja katalizira reakciju pretvorbe dopamina u noradrenalin. Aktivnost enzima u plazmi je stabilna karakteristika svakog pojedinca, a za njegovu aktivnost do 50 % odgovoran je funkcionalni polimorfizam (-1021CT) DBH lokusa. Cilj rada bio je usporediti polimorfizam - 1021CT DBH lokusa u zdravih osoba i oboljelih od PTSP, kako bi se ustanovilo da li se genetske promjene mogu povezati s pojavom PTSP. Izolirana je DNA 40 muških osoba oboljelih od PTSP i 40 zdravih muškaraca, koja je podvrgnuta PCR-RFLP analizi, te je utvrđena distribucija alela i genotipova za DBH. Rezultati su pokazali da učestalost recesivnog - 1021T alela u skupini oboljelih od PTSP iznosi 0.25, a u kontrolnoj skupini 0.26, te da ne postoji statistički značajna razlika (χ ² ; ; = 0.581, p= 0.748) između skupina. Na temelju usporedbe učestalosti i distribucije alela i genotipova između zdravih osoba i oboljelih od PTSP-a, nije moguće zaključiti o povezanosti između pojave PTSP i polimorfizma (-1021CT) u promotoru gena za DBH.
