Abstract

La integridad de las membranas plasmática y acrosomal es de gran importancia para la funcionalidad de los espermatozoides. En la actualidad existen varias pruebas para evaluar la integridad y/o funcionalidad de la membrana plasmática, entre ellas las tinciones vitales con fluorocromos (como por ejemplo la tinción con Diacetato de 6-carboxifluoresceína e Ioduro de Propidio) y la prueba de endósmosis o HOS test, ambas utilizadas de rutina en laboratorios de investigación para evaluar semen de llama (Giuliano et al, 2008; Carretero et al., 2012; Casaretto et al., 2012). En el semen de camélidos sudamericanos no se puede evaluar el acrosoma mediante microscopía de contraste de fase, ya que no se distingue como en otras especies. Por lo tanto diversas técnicas se han utilizado para poder evaluarlo utilizando tanto microscopía de campo claro como microscopía de fluorescencia.

The plasma and acrosomal membrane integrity is very important for sperm function. There are currently several tests to evaluate plasma membrane integrity and/or funtionality, including the vital staining with fluorochromes (such as for example: the stain with 6-carboxyfluorescein diacetate (6 CFDA) and propidium iodide (PI) and endosmosis test or hypo-osmotic swelling (HOS) test, weapons routinely used in research laboratories to evaluate Llama semen (Giuliano et al, 2008; Carretero et al., 2012; Casaretto et al., 2012). In South American camelids semen cannot evaluate the acrosome by phase contrast microscopy same manner in other species. Hence, need assessment techniques use bright field microscopy or fluorescence microscopy.
