Bromochalcona e inhibidores de cisteín proteasas

Abstract

Las chalconas son los precursores biosintéticos de los flavonoides y estudios recientes han demostrado que presentan actividades citotóxicas en diferentes líneas celulares tumorales incluyendo neoplasias hematológicas. Nos hemos centrado en el esqueleto de la chalconas como molde para elucidar las relaciones estructura- citotoxicidad de compuestos efectivos frente a células de leucemia humana. Una bromochalcona específica presentó mayor citotoxicidad frente a las células de leucemia humana que frente a células mononucleares de sangre periférica de voluntarios sanos. El efecto citotóxico de esta chalcona en células de leucemia es debido a la inducción de apoptosis. Los ejecutores fundamentales de este tipo de muerte celular son una familia de cisteín proteasas denominadas caspasas que se sintetizan como zimógenos y se activan por proteólisis. En esta comunicación presentamos el efecto de la combinación de la bromochalcona y los inhibidores de estas cisteín proteasas, tanto iniciadoras (caspasa-8 y -9) como ejecutoras (caspasa-3/7 y 6) con la finalidad de diseccionar las vías de señalización de activación de caspasas que están implicadas en la citotoxicidad. La muerte celular inducida por la bromochalcona en las células de leucemia humana U-937 es bloqueada por un inhibidor general de estas cisteín proteasas (z-VAD-fmk) y también por el inhibidor específico de la caspasa-8 (z-IETD-fmk) pero no por el inhibidor de la caspasa-9 (z-LEHD-fmk) indicando un papel clave de la vía extrínseca de inducción de apoptosis.
