Aislamiento y termoestabilidad de inhibidores de tripsina del frijol negro (P. vulgaris)

Abstract

En el presente trabajo se aislaron los inhibidores de tripsina del frijol negro (P. vulgaris) y se evaluó su estabilidad térmica. Los inhibidores fueron aislados por cromatografía de afinidad, obteniéndose, para los dos ensayos realizados, 0,0020 y 0,0029 g de inhibidos, con factores de purificación de 71.42 y 67.61, respectivamente. El producto obtenido fue sometido a electroforesis, determinándose que casi la totalidad de las proteínas presentes en el extracto original que no tenían actividad antitríptica, habían sido removidas en el producto aislado. Sin embargo, el patrón electroforético del producto obtenido no coincidió con el del extracto original. Para evaluar la estabilidad térmica se compararon las bandas electroforéticas obtenidas a partir de frijoles cocidos a presión atmosférica a 0, 30, 60, 90 y 120 min. Paralelamente se efectuó un ensayo de actividad antitríptica para las mismas muestras de frijol. Con ambos métodos, el electroforético (cualitativo) y el de competencia (cuantitativo) se determinó que a los 30 min. de cocción los inhibidores de tripsina habían sido ya destruidos. Del trabajo realizado se concluyó que los inhibidores de tripsina del frijol negro (P. vulgaris) son múltiples, aislándose 7 bandas con comportamiento electroforético distinto y que los inhibidores de tripsina son inestables a un tratamiento de cocción a presión atmosférica a partir de 30 min.
