Abstract

Bunga kecombrang (Nicolaia speciosa) memiliki kandungan kimia alkaloid, triterpenoid, steroid, flavonoid, dan polifenol yang memiliki aktivitas antibakteri. Penelitian ini bertujuan untuk memformulasikan ekstrak etil asetat bunga kecombrang dengan basis HPMC menjadi sediaan hidrogel yang stabil secara fisik dan melihat aktivitasnya terhadap bakteri Staphylococcus aureus. Bunga kecombrang diekstraksi secara maserasi bertingkat menggunakan pelarut n-heksan dan etil asetat. Gel ekstrak bunga kecombrang dibuat dengan variasi konsentrasi HPMC 3%, 5%, 8%, dan 10% b/b. Formula terbaik yang memenuhi syarat evaluasi uji sifat fisik dan stabil kemudian diuji aktivitas antibakterinya terhadap Staphylococcus aureus menggunakan metode difusi sumuran. Hasil penelitian menunjukkan bahwa perbedaan konsentrasi HPMC dapat mempengaruhi sifat fisik dan kestabilan hidrogel. Formula 1 adalah terbaik yang memenuhi syarat sifat fisik dan stabilitas sediaan dengan pH 5, viskositas 265.900-383.400 cps, daya sebar 4,67-5,20 cm, dan daya lekat 4,01-11,31 detik. Formula 1 menunjukkan aktivitas antibakteri dengan zona hambat sebesar 3,89 mm.
