Profil Senyawa Daidzein Umbi Bengkuang Serta Potensinya pada Tikus Putih Betina

Abstract

Penelitian ini bertujuan untuk menganalisis dinamika senyawa berbasis bahan aktif daidzein dalam umbi bengkuang serta potensinya pada uterus dan tulang.Penelitian menggunakan pendekatan eksperimen pola Rancangan Acak Lengkap dengan perlakuan pemberian air perasan umbi bengkuang dan senyawa sintetis daidzein pada 24 ekor tikus putih jenis Sprague Dawley umur 5 bulan, bobot badan 170-220 gram, selama 24 hari. Hewan coba dikelompokkan menjadi 3 kelompok perlakuan kontrol pemberian senyawa sintetis daidzein, dan pemberian air perasan umbi bengkuang, masing-masing hewan coba dilakukan pengulangan 8 kali. Takaran daidzein diberikan berdasarkan kadar daidzein pada 1,5 ml perasan umbi bengkuang. Pengambilan darah dilakukan pada 3 fraksi (jam ke-8, 16, dan 24 setelah perlakuan). Pembedahan dan pengambilan organ uterus dan tulang dilakukan hari ke-25, pembuatan preparat jaringan dengan pewarnaan HE. Hasil analisis HPLC kadar daidzein umbi bengkuang 108,831mg/100 g, kadar daidzein dalam serum darah pada 3 fraksi masing-masing adalah 96138,371 pg/1000 g, 130413,443 pg/1000 g dan 165991,504 pg/1000 g. Retensi daidzein umbi bengkuang dalam organ pada 3 fraksi masing-masing adalah 302819364,7 pg/ekor, 302818702,3 pg/ekor, dan 302818019,3 pg/ekor. Uji Oneway Anova jaringan miometrium dan osteoblas nilai P < 0,05. Kesimpulan menunjukkan bahwa umbi bengkuang mengandung senyawa daidzein mirip estrogen yang berpengaruh terhadap miometrium, tulang, dan serum.
