Uz modeli bāzēta biomasas iznākuma optimizācija un kontrole E. coli BL21 (DE3) hepatīta B-korantigēna (HBcAg) fermentācijas procesam ar piebarošanu

Abstract

Promocijas darba ietvaros, izmantojot matemātisko modelēšanu, tika noteikti dažāda veida un trajektorijas (substrāta) piebarošanas profili rekombinanta Escherichia coli BL21 (DE3) pBR327 hepatīta B kor-antigēna producenta biomasas ieguves fermentācijas procesam. Iegūtie piebarošanas profili paredzēti dažāda biomasas augšanas ātruma un iznākuma ieguvei dotajam procesam. Darba ietvaros ieviestas un aprobētas iepriekšnoteikto piebarošanas profilu izmeklētas piebarošanas ātruma tiešsaistes kontroles metodes – kontrole pēc pO2, un uz modeli bāzēta kontrole ar prognozi, kur abas metodes nodrošina augstāku procesa drošību un atkārtojamību, kas vēlamas GMP tipa procesiem. Darba izstrādes laikā tika realizēti priekš-eksperimenti kolbās, fizikālķīmisko parametru temperatūras un pH ietekmes noteikšanai uz biomasas iznākumu, kā arī veikti biomasas mērījuma precizitātes un kļūdu aprēķini. Piebarošanas profilu aprēķinam tika veikta fermentācijas procesu modelēšana, sākotnēji lietojot kolbu eksperimentu datus, tad modelējot fermentācijas procesus ar piebarošanu 5 l bioreaktora procesam. Tika identificēti 2 dažādas sarežģītības fermentācijas procesa modeļi kolbu un bioreaktora mērogiem, kur sarežģītākā modeļa gadījumā tika ņemta vērā acetātu uzkrāšanās. Identificētais vienkāršotais matemātiskais modelis, kurā netiek ņemta vērā acetātu ietekme, tika lietots uz modeli bāzētai piebarošanas ātruma kontrolei ar prognozi, realizācijai. Literatūras apskatā apkopota un izvērtēta informācija par laika periodu no 1949. gada līdz 2016. gadam
