Optimization of a protocol to obtain friable calli of borojó (Borojoa patinoi Cuatr.) phase I.

The influence of growth regulators to obtain friable calli in borojo (Borojoa patinoi Cuatr) was investigated. The used vegetable material was obtained from productive trees located in the town of San Luis, Antioquia, Colombia. Seeds were disinfected with 3% sodium hypochlorite for 20 minutes, plus 70% ethanol for 15 minutes with the addition of two tween 20 drops. In the different experiments, young leaves obtained in vitro from seeds were used as explants. They were cultivated in the MS medium supplemented with 2,4-D and combined with BAP and TDZ. No positive response related to the obtaining of friable callus was shown between the days 15 to 45. However, another test was carried out with the same explants in the MS medium with the addition of 1mg/L 2,4-D, as a medium for the embryogenesis initiation, 45 days later the formation of green yellow friable calli was observed. This work is a partial report of a macro project about the optimization of a protocol to obtain secondary metabolites of commercial interest trough cellular suspensions of Borojoa patinoi.
