Преодоление гепатотоксичности метотрексата: роль тритерпеноидов

Abstract

Проблема гепатотоксичности лекарственных средств, используемых в химиотерапии солидных и гематологических опухолей, остается актуальной. В обзоре приведен анализ молекулярных механизмов и клинических проявлений структурно-функциональных нарушений в печени при проведении химиотерапии. Особое внимание уделено изменениям, вызываемым метотрексатом — одним из базовых противоопухолевых препаратов в онкогематологии. Обсуждается возможность ограничения гепатотоксичности метотрексата с помощью природных соединений — тритерпеноидов. Приводятся данные о спектре их биологической активности, включая противоопухолевый, противовоспалительный, иммунотропный, антиоксидативный и органопротективный эффекты. Милиацин (3-b-метокси-D18-олеанен) — тритерпеноид растительного происхождения — рассматривается как перспективное средство для защиты печени от токсического воздействия метотрексата. Милиацин не изменяет фармакокинетику и противоопухолевую активность метотрексата.
