- In vivo monitoring of 5-FU

The aim of the present study was to establish the in vivo-monitoring of the enrichment of 5-FU <br> liposomes in liver and liver metastases by MRI methods. Relative signal intensities of liver and tumor <br> tissue were determined. After sacrifying animals concentrations of 5-FU and it´s active intracellular <br> metabolite M3 were measured by HPLC techniques. We used CC531 adenocarcinoma in the liver of <br> WAG/Rij rats as a standardized liver tumor model in our investigations. In a complete new aproach the <br> cytostatic agent 5-FU was encapsulated in PEG liposomes together with Gadolinium DTPA, a <br> commonly used contrast agent in MRI. We compared the systemic supply via the tail vein with the <br> locoregional application into the liver artery with or without embolizate Spherex and different control <br> groups. The results show changes in the pharmacokinetics of 5-FU liposomes caused by the binding of <br> Gd DTPA. Both contrast agent and cytostatic drug influence each other and led to a different <br> pharmacokinetic. The described method may improve the prognostic value of an antitumoral treatment <br> as well as to help to reduce the number of animals to be sacrifeced in pharmacoligical studies.<br>
