The Presence of a Cytochrome P450-like Protein in the Human Intestinal Micro┬Ć ora Eubacterium aerofaciens

Eighteen human intestinal microbial strains were tested for the presence of cytochrome P450, an enzyme involved in detoxifying and toxifying various xenobiotics. The polymerase chain reaction (PCR) technique was used to amplify conserved regions within the gene. From the human intestinal micro ora Eubacterium aerofaciens, a DNA fragment of the expected size could be ampli. ed, for which a percentage homology of 20% was determined. Southern blot analysis, using Pseudomonas putida P450 CAM probes, showed reactions with probes at 45 and 55°C. The carbon monoxide (CO) difference spectrum produced a P450-like pro. le for E. aerofaciens, similar to P. putida P450 CAM. The presence of cytochrome P450 in human intestinal  ora may in uence the expression of hepatic cytochrome P450s.Keywords : human intestinal  ora, cytochrome P450, PCR, CO difference spectrum.
