This proposal is based on our work over the last few years. Specifically: (1) Understanding the generation of the calcium signal has provided insights in how this second messenger works. We have shown: Local and global calcium signals: It has been a long-standaing problem to understand how a ubiquitous second messenger, like calcium, might be able to selectively activate processes inside cells. We showed that different agonists were capable of generating distinct patterns of calcium signal (Thorn and Petersen 1992).
