    Epigenetic marks are fundamental to normal development, but little is known about signals that dictate their placement. Insights have been provided by studies of imprinted loci in mammals, where monoallelic expression is epigenetically controlled. Imprinted expression is regulated by DNA methylation programmed during gametogenesis in a sex-specific manner and maintained after fertilization. At Rasgrf1 in mouse, paternal-specific DNA methylation on a differential methylation domain (DMD) requires downstream tandem repeats. The DMD and repeats constitute a binary switch regulating paternal-specific expression. Here, we define sequences sufficient for imprinted methylation using two transgenic mouse lines: One carries the entire Rasgrf1 cluster (RC); the second carries only the DMD and repeats (DR) from Rasgrf1. The RC transgene recapitulated all aspects of imprinting seen at the endogenous locus. DR underwent proper DNA methylation establishment in sperm and erasure in oocytes, indicating the DMD and repeats are sufficient to program imprinted DNA methylation in germlines. Both transgenes produce a DMD-spanning pit-RNA, previously shown to be necessary for imprinted DNA methylation at the endogenous locus. We show that when pit-RNA expression is controlled by the repeats, it regulates DNA methylation in cis only and not in trans. Interestingly, pedigree history dictated whether established DR methylation patterns were maintained after fertilization. When DR was paternally transmitted followed by maternal transmission, the unmethylated state that was properly established in the female germlines could not be maintained. This provides a model for transgenerational epigenetic inheritance in mice.

    Automatic news categorization is essential to automatically handle the classification of multi-label news articles in online portal. This research employs some potential methods to improve performance of hierarchical multi-label classifier for Indonesian news article. First potential method is using Convolutional Neural Network (CNN) to build the top level classifier. The second method could improve the classification performance by calculating the average of the word vectors obtained from distributed semantic model. The third method combines lexical and semantic method to extract documents features, which multiplied word term frequency (lexical) with word vector average (semantic). Model build using Calibrated Label Ranking as multi-label classification method, and trained using Naïve Bayes algorithm has the best F1-measure of 0.7531. Multiplication of word term frequency and the average of word vectors were also used to build this classifiers. This configuration improved multi-label classification performance by 4.25%, compared to the baseline. The distributed semantic model that gave best performance in this experiment obtained from 300-dimension word2vec of Wikipedia’s articles. The multi-label classification model performance is also influenced by news’ released date. The difference period between training and testing data would also decrease models’ performance.

    The early onset of covid-19 in 2020 causes a complete shutdown of all activities including those of services in the library to avoid direct contact with users in order to prevent the spread of this covid-19 virus. The purpose of this study is to fulfill the tasks that teachers have given you and to find out what innovations the unpad library has made to serve students or other users in spite of this covid-19 pandemic. The study USES descriptive qualitative methods on concepts related to library science, as well as to analyze the practices used in libraries during the pandemic. Data obtained through interviews conducted by researchers by sending some questions through the email of the unpad central library staff. The data already obtained will be presented in a descriptive form, so it will outline the importance of providing online library services by applying digital predating concepts. By subscribing to multiple journals and e-book databases, information-related services are still needed to support student and lecturer research, in this case libraries can engage in inter-library cooperation.     Designing multimodal freight transport networks can facilitate the economic development of regions and countries as well as help to reduce negative environmental impacts. It is therefore crucial that such be undertaken in areas where more priority is given on road-based freight transport systems. This paper proposes a model for strategic transport planning, particularly in freight terminal development and interregional freight transport network design. The model determines a suitable set of actions from a number of possible actions, such as improving the existing infrastructure or establishing new roads, railways, sea links, and freight terminals. Modelling is undertaken within the framework of bilevel programming, where a multimodal multiclass user traffic assignment technique is incorporated within the lower-level problem, whilst the upper-level problem determines the best combination of actions such that the freight-related benefit-cost ratio is maximised. The upper-level problem involves combinatorial optimisation, and a heuristic approach based on genetic local search is applied as a solution technique. Empirical results of the model as applied to an actual large-sized interregional intermodal freight transport network show that genetic local search could provide better performance as compared to other genetic algorithm-based, as well as tabu search-based, heuristics. The model is successfully applied to transport network planning in the Philippines, where the development of a freight transport network is necessary to increase the utilisation of other modes rather than road-based vehicles.

    In actualizing the Regional Government Budget ( APBD ) as an instrumen t for the desired social changes and in accordance to the purpose for decentralization, there needs to be the involvement of the civil society, community group s , and other stakeholders in public management to influence the policy formulation process and policy implementation which could improve public services and public accountability. The purpose of the current study was to describe the process of formulating the budgeting policy and identifying and plotting the participation of a number of actor s , both formal actor s and informal actor s, in the process of formulating the budgeting policy in the Banten Province Regional Government. The r esearch method employed was a qualitative research method using the NVivo 12 Pro software application . A number of the current study ’s findings which were related to the budgeting process were clearly linked to the actors’ political relation which would give rise to demands and requests in the form of actvity proposals . This process would also give rise to various compromise s and bargaining between the actors involved , including the value system which influences it . Moreover, the process of formulating the budgeting policy in Banten Province Regional Government in 2018, both in planning and budgeting , were done only as a procedure but not yet substantively . Therefore, budgeting policy formulation process needs to prioritize more widespread public participation, especially in discussions and collecting input or information for the government, both directly or indirectly so that a transparent and accountable process is created .

    The using of composits material currently more developed together with the using of it from the simpler like larges of housing tools to industry sector.The composite resech supported by fiber of eichhornia crassipes solms that has filler of cassava waste that has functions to know tensile strength and impact strength.This reseach used the various added of cassava waste , 0, 20, 25, and 30 (% of weight) and using of polyester resin matrix, the form of the composite speciment is plat with the fractions volume eichhornia crassipes solms fiber 20 % (constant), the fiber length 25 mm, because methode to found eichhornia crassipes solms fiber using methode of manual so that nothing of fibers have cuality and length with the same.Composite manufacturing process carried out by the methode of hand lay up with emphasis manually using glass as a mold and pressure.The results tensile reseach founded that lests of filler as used so that the higest that tensile strength, the highest tensile strength at variations added of cassava waste 0 % ( 9,311 N/mm2 ) and for impact strength the biggest added of cassava waste so that the highest energy to fight of materials , the high tensile strength found in the variations added of cassava waste 25 % ( 0,018282 J/mm2 ).

    This research aimed to study the substitution of soybean meal (SBM) with Indigofera zollingeriana top leaf meal (ILM) in the diet on egg quality of Japanese quails. The experiment used a completely randomized design with five treatments and four replications (ten quails of each replication). The dietary treatment contained five combination of SBM and ILM, R0= diets contained 18% SBM without ILM, R1= diet contained 16.2% SBM and 2.66% ILM, R2= diet contained 14.4% SBM and 5.32% ILM, R3= diet contained 12.6% SBM and 7.98% ILM, R4= diet contained 9% SBM and 13.3% ILM. The results showed that the use of 13.3% ILM (R4) significantly (P<0.05) increased feed consumption, egg weight, yolk colour score, egg cholesterol, and reduced malondialdehyde level. The conclusion of this study was I. zollingeriana top leaf meal could be used as much as 13.3% in the diets. The use of I. zollingeriana top leaf meal could improve the quality of eggs physically and chemically. Key words: eggs quality, Indigofera zollingeriana, soybean meal, quail
