    Electron energy loss spectroscopy (EELS) has become a mainstream characterization method. It provides access to high spatial resolution analytical and electronic properties information in a TEM. When combined with the imaging and diffraction capabilities of a TEM a nanomaterial can be characterized in great detail. The aim is to review the principles and implementation of angle- and spatially-resolved EELS and its applications to study of material properties at nanoscale.     Leukocyte recruitment towards sites of infection involves coordinated ligand-receptor interactions between leukocytes and the vascular wall. Selectins transiently bind to selectin ligands to enable leukocyte tethering and rolling on the vascular wall prior to leukocyte adhesion and transmigration into tissues. These interactions occur following post-translational modifications of selectin ligands, including terminal fucosylation by Fucosyltransferase-IV and -VII. In mice, deficiency in both enzymes (FtDKO) leads to loss of leukocyte tethering and rolling, increased circulating neutrophils and impaired T cell trafficking. However, the role of functional selectin ligand interactions in modulating immunity to bacterial infections remains unclear. We analyzed the kinetics of infection and modulation of immune responses in FtDKO mice infected with Listeria monocytogenes, an intracellular bacterium. FtDKO mice controlled systemic infection more rapidly relative to wild-type mice. This was correlated to increased number and functionality of neutrophils. Adoptive transfer of bone marrow cells from FtDKO mice into irradiated wild-type mice resulted in expansion of neutrophil populations and increased resistance to infection in a neutrophil-dependent manner. Functional selectin ligand deficiency on neutrophils was correlated to increased resistance to cell death. Thus, functional selectin ligand expression is a potential target for enhancing innate immunity against bacterial infection.     Zernike-like phase plate contrast can be obtained in a transmission electron microscope by introducing a phase shift between direct and diffracted beams in the back focal plane of objective lens. A particularly convenient implemenation utilizes a uniform thin film with its contrast transfer properties determined by local modifications of the film by the primary electron beam. The need for accurate positioning of the device in back focal plane is eliminated because the phase shifting patch size and position is determined by the primary beam size and position. Such hole free phase plate can increase image contrast by a factor of two to four. While hole free phase plate contrast can not be quantitatively interpreted at present, it can be used to image for example magnetic field inside sample and fringing field in vacuum near sample. It can be also applied to imaging of biological materials. The origin of the hole free phase plate phase shift is not fully understood.     Cold spray offers unique possibilities in processing intermetallic-forming or phase-segregating metallic alloys, which are difficult to address by classical melt-metallurgical processes such as casting or thermal spraying due to the formation of large brittle regions or low melting point and volatile eutectics. An example is the production of thick coatings of the copper-indium-gallium (CIG) system on plates or tubes, as they are required in the fabrication of the CIGS(Se) absorber layer of thin film photovoltaic solar modules on an industrial-scale. At the relevant compositions, extended solidification intervals, such as in casting, create material segregation into large brittle CuGa2-type regions intermixed in an indium-rich matrix rendering the cast non-uniform and fragile with high internal stresses. Shrinkage effect further promote porosity. The equilibrium phase diagrams even allow the retention of pure Ga and the In-Ga eutectic, essentially liquid materials at room temperature and highly undesirable for the application. On the other hand, using rapid solidification by gas atomizing the Cu-In-Ga to produce cold spray powder promotes a fine-scaled and uniform phase distribution and microstructure of Cu(In,Ga), CuIn and In. By decoupling the material synthesis from the forming process, this phase distribution can be fully preserved in the cold spray deposition process to advantageously fabricate the coatings for that application.     Electron energy loss spectroscopy (EELS) has become a mainstream characterization method. It provides access to high spatial resolution analytical and electronic properties information in a TEM. When combined with the imaging and diffraction capabilities of a TEM a nanomaterial can be characterized in great detail. The aim is to review the principles and implementation of angle- and spatially-resolved EELS and its applications to study of material properties at nanoscale.     Leukocyte recruitment towards sites of infection involves coordinated ligand-receptor interactions between leukocytes and the vascular wall. Selectins transiently bind to selectin ligands to enable leukocyte tethering and rolling on the vascular wall prior to leukocyte adhesion and transmigration into tissues. These interactions occur following post-translational modifications of selectin ligands, including terminal fucosylation by Fucosyltransferase-IV and -VII. In mice, deficiency in both enzymes (FtDKO) leads to loss of leukocyte tethering and rolling, increased circulating neutrophils and impaired T cell trafficking. However, the role of functional selectin ligand interactions in modulating immunity to bacterial infections remains unclear. We analyzed the kinetics of infection and modulation of immune responses in FtDKO mice infected with Listeria monocytogenes, an intracellular bacterium. FtDKO mice controlled systemic infection more rapidly relative to wild-type mice. This was correlated to increased number and functionality of neutrophils. Adoptive transfer of bone marrow cells from FtDKO mice into irradiated wild-type mice resulted in expansion of neutrophil populations and increased resistance to infection in a neutrophil-dependent manner. Functional selectin ligand deficiency on neutrophils was correlated to increased resistance to cell death. Thus, functional selectin ligand expression is a potential target for enhancing innate immunity against bacterial infection.     Zernike-like phase plate contrast can be obtained in a transmission electron microscope by introducing a phase shift between direct and diffracted beams in the back focal plane of objective lens. A particularly convenient implemenation utilizes a uniform thin film with its contrast transfer properties determined by local modifications of the film by the primary electron beam. The need for accurate positioning of the device in back focal plane is eliminated because the phase shifting patch size and position is determined by the primary beam size and position. Such hole free phase plate can increase image contrast by a factor of two to four. While hole free phase plate contrast can not be quantitatively interpreted at present, it can be used to image for example magnetic field inside sample and fringing field in vacuum near sample. It can be also applied to imaging of biological materials. The origin of the hole free phase plate phase shift is not fully understood.     Cold spray offers unique possibilities in processing intermetallic-forming or phase-segregating metallic alloys, which are difficult to address by classical melt-metallurgical processes such as casting or thermal spraying due to the formation of large brittle regions or low melting point and volatile eutectics. An example is the production of thick coatings of the copper-indium-gallium (CIG) system on plates or tubes, as they are required in the fabrication of the CIGS(Se) absorber layer of thin film photovoltaic solar modules on an industrial-scale. At the relevant compositions, extended solidification intervals, such as in casting, create material segregation into large brittle CuGa2-type regions intermixed in an indium-rich matrix rendering the cast non-uniform and fragile with high internal stresses. Shrinkage effect further promote porosity. The equilibrium phase diagrams even allow the retention of pure Ga and the In-Ga eutectic, essentially liquid materials at room temperature and highly undesirable for the application. On the other hand, using rapid solidification by gas atomizing the Cu-In-Ga to produce cold spray powder promotes a fine-scaled and uniform phase distribution and microstructure of Cu(In,Ga), CuIn and In.
