Obiecującą perspektywą dla skuteczności leczenia nowotworów jest łączenie klasycznej chemioterapii z terapią ukierunkowaną na różne cele molekularne. Zastosowanie inhibitorów białek zaangażowanych w ścieżki sygnalizacyjne, związane z powstawaniem i rozwojem nowotworu, może wpłynąć na skuteczność i specyficzność stosowanych leków. Fosfataza DUSP6 jest białkiem, którego rola w procesie nowotworzenia jest niejednoznaczna. Wskazuje się zarówno na jej właściwości supresyjne, jak i onkogenne. Celem projektu badawczego była ocena wpływu inhibitora fosfatazy DUSP6 (BCI) na stopień cytotoksyczności onkoterapeutyków – irinotekanu i oksaliplatyny w komórkach raka jelita grubego, w układzie in vitro. W badaniu wykorzystano dwie ludzkie linie komórkowe, różniące się statusem genu BRCA2, a tym samym zdolnością do naprawy uszkodzeń DNA – DLD1 oraz DLD1 BRCA2 (-/-). Układy badawcze rozszerzono o zastosowanie inhibitora kinazy serynowo-treoninowej ATM (KU60019), będącej jednym z kluczowych białek mechanizmu naprawy DNA. W pierwszym etapie badań, dokonano oceny cytotoksyczności (test MTT) pojedynczych związków, a następnie ich układów. Oceniono wpływ wybranych stężeń BCI na właściwości chemioterapeutyków, również w obecności inhibitora ATM. W kolejnym etapie, wybrane układy doświadczalne poddano analizie cytometrycznej (Alexa Fluor®488 Annexin V/Dead Cell Apoptosis Kit), celem sprawdzenia ich aktywności w badanych komórkach oraz wskazania, jakiego rodzaju śmierć komórkową indukują. Combining traditional chemotherapy with treatments that focus on different molecular targets is a useful approach to enhance the effectiveness of cancer treatments. The efficiency and specificity of drugs can be affected by the use of inhibitors of proteins that are involved in signaling pathways associated with cancer development. DUSP6 is a phosphatase protein with an ambiguous role in tumorigenesis. It was found to have both suppressive and oncogenic properties. This research project aimed to evaluate the effect of the phosphatase DUSP6 inhibitor (BCI) on the cytotoxicity of the therapeutical agents used in cancer treatment (irinotecan and oxaliplatin) in colorectal cancer cells in vitro. Two cell lines differing in BRCA2 gene status and DNA repair capacity were used in the study – DLD1 and DLD1 BRCA2 (-/-). The research was extended with the use of ATM kinase inhibitor (KU60019), one of the key proteins involved in cellular DNA repair mechanisms. The cytotoxicity of the individual compounds (MTT assay) was analyzed in the first stage of the study, followed by the investigation of the cytotoxicity of the compounds in combinations. The effect of selected concentrations of BCI on the properties of chemotherapeutics was assessed, also in the presence of an ATM inhibitor. In the next step, selected compound combinations were subjected to cytometric analysis (Alexa Fluor®488 Annexin V/Dead Cell Apoptosis Kit) to evaluate their activity in the studied cells and to indicate the type of cell death that they can induce. The file contains data related to materials published in 2022 The aim of this study is to investigate the effect of microplastics (MPs) on the biodegradation activity of microscopic soil fungi and fungal-plant interactions. Plastic mulch films are intensively used in agriculture to suppress weed growth, prevent moisture loss, and improve crop yield. On the other hand, urban sewage sludge containing microplastic particles is used to fertilize soil. These particles pollute agricultural soil and could affect the soil ecosystem. As a result of human activity agricultural soil can also contain heavy metals and pesticides. Fungi are an important component of terrestrial ecology. Many of them have a positive (e.g. Trichoderma spp.) or negative (e.g. Fusarium spp.) effect on plant health, which is of major importance for human economy. The planned research on the impact of plastics on the plant growth in the presence of fungi has an innovative character. In the present project we hypothesize that MPs affect the biodegradation activity of fungi and fungal-plant interactions. To test our hypothesis we will examine (1) whether fungal activities expressed as their biodegradation potential towards herbicides in heavy metals presence are modified in the environment polluted with MPs as compared to control settings, (2) whether microplastic particles have effect on parallel interactions between wheat (Triticum aestivum) and the fungi Trichoderma spp. and/or Fusarium spp.. We also hypothesize that microplastics, apart from affecting the interaction between fungi and wheat, might have an impact on plant growth, germination, chlorophyll content and accumulation of metals (Zn and Cu) and be bound by wheat itself. Moreover, we will use low density polyethylene (LDPE), which is most often found in soils as well as poly-butyleneadipate-co-terephthalate (PBAT) as potential biodegradable alternative of LDPE as future plastic mulch film materials. The effects of MPs on soil fungi remain largely unexplored and represent a relevant area for future research. The data obtained will expand our knowledge of the influence of MPs on terrestrial microorganisms and their effects on plants. Obiecującą perspektywą dla skuteczności leczenia nowotworów jest łączenie klasycznej chemioterapii z terapią ukierunkowaną na różne cele molekularne. Zastosowanie inhibitorów białek zaangażowanych w ścieżki sygnalizacyjne, związane z powstawaniem i rozwojem nowotworu, może wpłynąć na skuteczność i specyficzność stosowanych leków. Fosfataza DUSP6 jest białkiem, którego rola w procesie nowotworzenia jest niejednoznaczna. Wskazuje się zarówno na jej właściwości supresyjne, jak i onkogenne. Celem projektu badawczego była ocena wpływu inhibitora fosfatazy DUSP6 (BCI) na stopień cytotoksyczności onkoterapeutyków – irinotekanu i oksaliplatyny w komórkach raka jelita grubego, w układzie in vitro. W badaniu wykorzystano dwie ludzkie linie komórkowe, różniące się statusem genu BRCA2, a tym samym zdolnością do naprawy uszkodzeń DNA – DLD1 oraz DLD1 BRCA2 (-/-). Układy badawcze rozszerzono o zastosowanie inhibitora kinazy serynowo-treoninowej ATM (KU60019), będącej jednym z kluczowych białek mechanizmu naprawy DNA. W pierwszym etapie badań, dokonano oceny cytotoksyczności (test MTT) pojedynczych związków, a następnie ich układów. Oceniono wpływ wybranych stężeń BCI na właściwości chemioterapeutyków, również w obecności inhibitora ATM. W kolejnym etapie, wybrane układy doświadczalne poddano analizie cytometrycznej (Alexa Fluor®488 Annexin V/Dead Cell Apoptosis Kit), celem sprawdzenia ich aktywności w badanych komórkach oraz wskazania, jakiego rodzaju śmierć komórkową indukują. Combining traditional chemotherapy with treatments that focus on different molecular targets is a useful approach to enhance the effectiveness of cancer treatments. The efficiency and specificity of drugs can be affected by the use of inhibitors of proteins that are involved in signaling pathways associated with cancer development. DUSP6 is a phosphatase protein with an ambiguous role in tumorigenesis. It was found to have both suppressive and oncogenic properties. This research project aimed to evaluate the effect of the phosphatase DUSP6 inhibitor (BCI) on the cytotoxicity of the therapeutical agents used in cancer treatment (irinotecan and oxaliplatin) in colorectal cancer cells in vitro. Two cell lines differing in BRCA2 gene status and DNA repair capacity were used in the study – DLD1 and DLD1 BRCA2 (-/-). The research was extended with the use of ATM kinase inhibitor (KU60019), one of the key proteins involved in cellular DNA repair mechanisms. The cytotoxicity of the individual compounds (MTT assay) was analyzed in the first stage of the study, followed by the investigation of the cytotoxicity of the compounds in combinations. The effect of selected concentrations of BCI on the properties of chemotherapeutics was assessed, also in the presence of an ATM inhibitor. In the next step, selected compound combinations were subjected to cytometric analysis (Alexa Fluor®488 Annexin V/Dead Cell Apoptosis Kit) to evaluate their activity in the studied cells and to indicate the type of cell death that they can induce. The file contains data related to materials published in 2022 The aim of this study is to investigate the effect of microplastics (MPs) on the biodegradation activity of microscopic soil fungi and fungal-plant interactions. Plastic mulch films are intensively used in agriculture to suppress weed growth, prevent moisture loss, and improve crop yield. On the other hand, urban sewage sludge containing microplastic particles is used to fertilize soil. These particles pollute agricultural soil and could affect the soil ecosystem. As a result of human activity agricultural soil can also contain heavy metals and pesticides. Fungi are an important component of terrestrial ecology. Many of them have a positive (e.g. Trichoderma spp.) or negative (e.g. Fusarium spp.) effect on plant health, which is of major importance for human economy. The planned research on the impact of plastics on the plant growth in the presence of fungi has an innovative character. In the present project we hypothesize that MPs affect the biodegradation activity of fungi and fungal-plant interactions. To test our hypothesis we will examine (1) whether fungal activities expressed as their biodegradation potential towards herbicides in heavy metals presence are modified in the environment polluted with MPs as compared to control settings, (2) whether microplastic particles have effect on parallel interactions between wheat (Triticum aestivum) and the fungi Trichoderma spp. and/or Fusarium spp.. We also hypothesize that microplastics, apart from affecting the interaction between fungi and wheat, might have an impact on plant growth, germination, chlorophyll content and accumulation of metals (Zn and Cu) and be bound by wheat itself. Moreover, we will use low density polyethylene (LDPE), which is most often found in soils as well as poly-butyleneadipate-co-terephthalate (PBAT) as potential biodegradable alternative of LDPE as future plastic mulch film materials. The effects of MPs on soil fungi remain largely unexplored and represent a relevant area for future research. The data obtained will expand our knowledge of the influence of MPs on terrestrial microorganisms and their effects on plants.
