With this one sentence, Christopher Morley (1890–1957) beautifully summarized the unique relationship of dogs with humans, and prompts the questions that have interested many researchers, both then and now. When we talk to dogs, are we understood? And, if so, how flexible is the understanding of a dog of our communication? And what is it about dogs that makes them so attached to us? Has domestication changed dogs in such a way that they can develop this special relationship with us? And how exactly has the domestication process influenced dog behaviour? What changed from wolf to dog to make dogs such valuable companions to man that wherever there are humans, there are dogs?

    In order to strengthen the interest of students in the field of programming, robots are increasingly being used in courses of study with information technology content. This article describes the use of the robot Vector, manufactured by Anki, in a controlled laboratory experiment with undergraduate students. The aim of the research was to investigate the influence of robots on the motivation of programming beginners from the perspective of the students. The data of the study was collected using the Intrinsic Motivation Inventory. The subsequent analysis was performed using descriptive statistics and Chi2-tests. In addition, a computer-aided content analysis as well as a factor analysis was used as a second evaluation method. The research results showed a consistently positive influence of the Vector robot on the motivational behavior of the students. In particular, significant dependencies between the female gender and individual subcategories of the Intrinsic Motivation Inventory could be demonstrated. The generalization of the research results is limited due to the small sample size and the exclusive participation of programming beginners.

    Die amyotrophe Lateralsklerose (ALS) ist eine neurodegenerative Erkrankung, die innerhalb von 3–5 Jahren zum Tode fuhrt. Die bisher einzige Substanz mit marginalem therapeutischem Potenzial ist der Glutamatantagonist Riluzol, der jedoch nur zu einer durchschnittlichen Lebensverlangerung von 3–4 Monaten fuhrt. Daher hat die symptomatische Therapie einen hohen Stellenwert. In zahlreichen tierexperimentellen und klinischen Studien werden potenziell neuroprotektive Wirkstoffe fur eine kausale Behandlung untersucht. In diesem Ubersichtsartikel sollen im Kontext mit den zugrunde liegenden Wirkmechanismen aktuelle klinischen ALS-Studien vorgestellt werden.

    Methods: Selectivity and activity were investigated in vitro using cell lines, peripheral blood mononuclear cells (PBMCs) or isolated plasmacytoid dendritic cells (pDCs). In vivo studies were also performed in cynomolgus monkeys orally administered single doses of 0.1 to 2.0mg/kg; daily doses of 0.1 to 1.0mg/kg for 7 days or every other day doses of 0.05 to 1.5mg/kg for 28 days. Assessment after intravenous infusion and in portal vein cannulated animals was also done. We evaluated levels of serum cytokines and RNA of select interferon stimulated genes (ISGs) in blood cells. Tolerability was evaluated by clinical observations and laboratory testing. Results: GS-9620 activated TLR7 with approximately 100-fold selectivity over TLR8. In vitro, GS-9620 induced interferon-alpha (IFN) and other cytokines in human and cynomolgus monkey PBMCs and/or pDCs with a minimum effective concentration ranging from 14 to 66nM in humans and with 5-fold less potency in monkeys. GS-9620 induced B and T cell activation as shown by increased expression of CD69, CD86 and HLA-DR on B lymphocytes in PBMCs, CD69 and HLA-DR on isolated B lymphocytes, and CD25 and CD69 in a small percentage of CD4+ or CD8+ T lymphocytes in PBMCs but not in isolated T lymphocytes. In cynomolgus monkeys, GS-9620 was well tolerated at the highest oral doses evaluated including 1.5mg/kg every other day for 28 days. Dose and exposure dependent increases in IFN-alpha, immunomodulatory cytokines, chemokines, and peripheral blood cell ISGs were noted. There was no evidence of tachyphylaxis following every other day dosing. Oral administration resulted in limited systemic bioavailability but high oral absorption. Conclusions: GS-9620 is a selective potent TLR7 agonist which elucidated a selective pattern of cytokines and activated B and T lymphocytes. GS-9620 was well tolerated in cynomolgus monkeys with every other day oral dosing for 28 days. This resulted in dose-dependent IFN-alpha and ISG responses with no evidence of tachyphylaxis.

    The increasing share of renewable energies requires the installation of large-scale electricity storage capacities in addition to grid expansion. Significant contribution to reach this goal is provided by adiabatic compressed air energy storage power plants (A-CAES), key elements in future electricity transmission systems. This technology allows efficient, local zero-emission electricity storage on the basis of compressed air in underground caverns in combination with thermal energy storage systems and, in contrast to pumped storage power plants (PSPP), it demands no overground geological requirements. Despite the achieved success of A-CAES systems in terms of efficiency and cost, further improvements in dynamics and flexibility are needed. One promising solution to fulfil these dynamic requirements is based on the integration of an additional power-to-heat element (P2H) operating during the charging periods. This modification allows increased power plant flexibility and further cost reductions due to increased thermal storage densities but is simultaneously associated with concept-dependent decreasing total round trip efficiencies. For the identification of suitable configurations, adequate concepts must be elaborated, and the influence on round trip efficiency as well as on cost reduction potential must be investigated. For this purpose, a system model for a two-stage A-CAES configuration is established and used for large simulation studies related to P2H locations and power, thermal energy storage systems, and central process variables. Therefore, time-efficient model reductions with well-justified assumptions are conducted, offering a simplified transient implementation of thermal energy options in the system simulation. On the basis of a promising P2H configuration including high potentials for cost reduction and moderate losses in round trip efficiency, an alternative concept is presented offering high exergetic utilization and additional cost reductions, which can be treated as a base for upgrading the existing CAES power plants and for modifying operational concepts.

    Ozone reacts slowly with Ag+ (circumneutral pH, k = (11 ± 3) × 10−2 M−1 s−1). After some time, ozone decay kinetics may suddenly become faster with the concomitant formation of silver sol. As primary process, an O-transfer from O3 to Ag(I) is suggested, whereby Ag(III) is formed [Ag+ + O3 + 2 H2O → Ag(OH)3 + O2 + H+]. This conproportionates with Ag(I), which is in large excess, leading to Ag(II) [Ag+ + Ag(OH)3 ⇄ 2 Ag(OH)+ + HO−]. Further, Ag(II) reacts with ozone in a high exergonic reaction [Ag(OH)+ + O3 → Ag + 2 O2 + H+], where ozone acts as a reducing agent. Thereby, a single silver atom, Ag, is formed that can be oxidized by O2 and O3 or can aggregate to a silver sol. Aggregation slows down the rate of oxidation. When Ag+ is complexed by acetate ions, ozone decay and silver sol formation are speeded up by enhancing Ag(II) formation [Ag(I)acetate + O3 → Ag(III)acetate → Ag(II) + CO2 + •CH3].     X-ray magnetic circular dichroism yields at inner-core absorption edges large values up to 50%. This serves as huge magnetic contrast mechanism to image magnetic domain structures. The current status of magnetic soft X-ray microscopy providing currently a resolution down to about 20nm to study static domain configurations is demonstrated by selected examples on high magnetostrictive thin films and laterally microstructured elements measured at the XM-I microscope at the ALS (Berkeley CA). Element-specificity allows insight into magnetic coupling mechanisms and the evolution of the domain patterns within full hysteresis loops can be studied by recording the images within external magnetic fields. Both in-plane and out-of-plane configurations of the magnetization can be addressed by tilting the sample at an axis perpendicular to the photon propagation direction. Magnetic X-PEEM measurements obtained at the Swiss Light Source provide complementary information. Spin dynamics, i.e. precession and relaxation of the magnetization, can be addressed by taking into account the inherent pulsed time structure of synchrotrons, First results with a sub-lOOpsec time resolution obtained in a stroboscopic pump-probe experiment with a patterned permalloy element at the XM-1 are reported.

    During the last decade, analytical methods for the detection and quantification of proteins and peptides in biological samples have been considerably improved. It is therefore now possible to compare simultaneously the expression levels of hundreds or thousands of proteins in different types of tissue, for example, normal and cancerous, or in different cell lines. In this chapter, we illustrate statistical designs for such proteomics experiments as well as methods for the analysis of resulting data. In particular, we focus on the preprocessing and analysis of protein expression levels recorded by the use of either two-dimensional gel electrophoresis or mass spectrometry.

    In order to strengthen the interest of students in the field of programming, robots are increasingly being used in courses of study with information technology content. This article describes the use of the robot Vector, manufactured by Anki, in a controlled laboratory experiment with undergraduate students. The aim of the research was to investigate the influence of robots on the motivation of programming beginners from the perspective of the students. The data of the study was collected using the Intrinsic Motivation Inventory. The subsequent analysis was performed using descriptive statistics and Chi2-tests. In addition, a computer-aided content analysis as well as a factor analysis was used as a second evaluation method. The research results showed a consistently positive influence of the Vector robot on the motivational behavior of the students. In particular, significant dependencies between the female gender and individual subcategories of the Intrinsic Motivation Inventory could be demonstrated. The generalization of the research results is limited due to the small sample size and the exclusive participation of programming beginners.

    Die amyotrophe Lateralsklerose (ALS) ist eine neurodegenerative Erkrankung, die innerhalb von 3–5 Jahren zum Tode fuhrt. Die bisher einzige Substanz mit marginalem therapeutischem Potenzial ist der Glutamatantagonist Riluzol, der jedoch nur zu einer durchschnittlichen Lebensverlangerung von 3–4 Monaten fuhrt. Daher hat die symptomatische Therapie einen hohen Stellenwert. In zahlreichen tierexperimentellen und klinischen Studien werden potenziell neuroprotektive Wirkstoffe fur eine kausale Behandlung untersucht. In diesem Ubersichtsartikel sollen im Kontext mit den zugrunde liegenden Wirkmechanismen aktuelle klinischen ALS-Studien vorgestellt werden.

    Methods: Selectivity and activity were investigated in vitro using cell lines, peripheral blood mononuclear cells (PBMCs) or isolated plasmacytoid dendritic cells (pDCs). In vivo studies were also performed in cynomolgus monkeys orally administered single doses of 0.1 to 2.0mg/kg; daily doses of 0.1 to 1.0mg/kg for 7 days or every other day doses of 0.05 to 1.5mg/kg for 28 days. Assessment after intravenous infusion and in portal vein cannulated animals was also done. We evaluated levels of serum cytokines and RNA of select interferon stimulated genes (ISGs) in blood cells. Tolerability was evaluated by clinical observations and laboratory testing. Results: GS-9620 activated TLR7 with approximately 100-fold selectivity over TLR8. In vitro, GS-9620 induced interferon-alpha (IFN) and other cytokines in human and cynomolgus monkey PBMCs and/or pDCs with a minimum effective concentration ranging from 14 to 66nM in humans and with 5-fold less potency in monkeys. GS-9620 induced B and T cell activation as shown by increased expression of CD69, CD86 and HLA-DR on B lymphocytes in PBMCs, CD69 and HLA-DR on isolated B lymphocytes, and CD25 and CD69 in a small percentage of CD4+ or CD8+ T lymphocytes in PBMCs but not in isolated T lymphocytes. In cynomolgus monkeys, GS-9620 was well tolerated at the highest oral doses evaluated including 1.5mg/kg every other day for 28 days. Dose and exposure dependent increases in IFN-alpha, immunomodulatory cytokines, chemokines, and peripheral blood cell ISGs were noted. There was no evidence of tachyphylaxis following every other day dosing. Oral administration resulted in limited systemic bioavailability but high oral absorption. Conclusions: GS-9620 is a selective potent TLR7 agonist which elucidated a selective pattern of cytokines and activated B and T lymphocytes. GS-9620 was well tolerated in cynomolgus monkeys with every other day oral dosing for 28 days. This resulted in dose-dependent IFN-alpha and ISG responses with no evidence of tachyphylaxis.
