Background: Intra-cellular and inter-cellular protein translocation can be observed by microscopic imaging of tissue sections prepared immunohistochemically. A manual densitometric analysis is time-consuming, subjective and error-prone. An automated quantification is faster, more reproducible, and should yield results comparable to manual evaluation. The automated method presented here was developed on rat liver tissue sections to study the translocation of bile salt transport proteins in hepatocytes. For validation, the cholestatic liver state was compared to the normal biological state.Results: An automated quantification method was developed to analyze the translocation of membrane proteins and evaluated in comparison to an established manual method. Firstly, regions of interest (membrane fragments) are identified in confocal microscopy images. Further, densitometric intensity profiles are extracted orthogonally to membrane fragments, following the direction from the plasma membrane to cytoplasm. Finally, several different quantitative descriptors were derived from the densitometric profiles and were compared regarding their statistical significance with respect to the transport protein distribution. Stable performance, robustness and reproducibility were tested using several independent experimental datasets. A fully automated workflow for the information extraction and statistical evaluation has been developed and produces robust results.Conclusions: New descriptors for the intensity distribution profiles were found to be more discriminative, i.e. more significant, than those used in previous research publications for the translocation quantification. The slow manual calculation can be substituted by the fast and unbiased automated method.

金沢大学医薬保健研究域医学系 Y染色体特異的マイナー組織適合抗原であるH-Yペプチドは男性にのみ発現し,HLA-A2に表出される.まず,HLA一致姉からの同種骨髄移植後に慢性骨髄性白血病リンパ性急性転化を来した男性患者(HLA-A2陽性)を対象に,患者の白血病細胞に対し高い細胞傷害活性を示すドナー由来のT細胞を作成した.H-Yペプチドでパルスしたドナー由来樹状細胞を抗原提示細胞に用いてドナーのT細胞を繰り返し刺激することにより,細胞傷害性T細胞を樹立した.この細胞傷害性T細胞は,ドナーの樹状細胞に対しパルスするペプチド濃度依存性の細胞傷害活性を示した.さらに,患者白血病細胞・線維芽細胞,男性リンパ芽球性リンパ細胞株細胞に対し,HLA-A2拘束性の細胞傷害活性を示した.次に,同じ男性患者が再移植を受けたのち分子遺伝学的寛解を維持していた時期の流血中に,インターフェロン(IFN)γ発現とT細胞レセプターを指標としてH-Yペプチド特異的T細胞を検出した.患者血液からCD8陽性細胞を分離し,H-Yペプチドで5時間刺激したところ,6.8%がIFN-γ陽性となった.このCD8陽性IFN-γ陽性細胞のT細胞レセプターβ鎖のCDR3領域をPCRにより増幅し,サイズ・スペクトラタイピングを行った.その結果,BV22においてin vitroで誘導された細胞傷害性T細胞と同じサイズのピークが検出された.塩基配列を決定したところ,患者の流血中に存在しているH-Yペプチド反応性T細胞とin vitroで樹立した細胞傷害性T細胞で共通のCDR3領域の配列が証明された.流血中に存在していたCD8陽性細胞は,H-Yペプチド特異的にIFN-γを発現することから,同種免疫による抗腫瘍効果の担当細胞であった可能性がある. 研究課題/領域番号:13770578, 研究期間(年度):2001-2002 出典:「INFγ発現とT細胞レセプターを指標としたマイナー組織適合抗原特異的T細胞の検出」研究成果報告書 課題番号13770578(KAKEN:科学研究費助成事業データベース(国立情報学研究所)) ( https://kaken.nii.ac.jp/ja/grant/KAKENHI-PROJECT-13770578/ )を加工して作成

Η εμβέλεια του ποζιτρονίου είναι ένας από τους κυριότερους παράγοντες περιορισμού της ανάλυσης της εικόνας στην Ποζιτρονική Τομογραφία (PET). Στην παρούσα ερευνητική εργασία μελετήθηκε η επίδραση του μαγνητικού πεδίου στην εμβέλεια του ποζιτρονίου σε διπλό σύστημα Ποζιτρονικής Τομογραφίας Εκπομπής και Μαγνητικού Τομογράφου (PET-MR) μέσω προσομοίωσης. Το ισχυρό μαγνητικό πεδίο του Μαγνητικού Τομογράφου δημιουργεί συστηματικά επηρεασμένα αποτελέσματα τα οποία διαφοροποιούνται από τα αντίστοιχα εκτός μαγνητικού πεδίου. Λόγω εμφάνισης δυνάμεων Lorentz στο εκπεμπόμενο ποζιτρόνιο αναμένεται μείωση της εμβέλειας του ποζιτρονίου στο κάθετο στο μαγνητικό πεδίο επίπεδο. Για τον έλεγχο της υπόθεσης αυτής αλλά και την ποσοτικοποίηση του αποτελέσματος, πραγματοποιήθηκαν Monte Carlo προσομοιώσεις βασισμένες στο λογισμικό GEANT4/GATE σημειακής β+ ραδιενεργού πηγής εντός απλού συστήματος PET, με εισαγωγή στατικού και ομογενούς μαγνητικού πεδίου. Για την ανακατασκευή της εικόνας χρησιμοποιήθηκαν αναλυτικοί αλγόριθμοι που έχουν αναπτυχθεί στο εργαστήριό μας (REC3D), οι οποίοι πραγματοποιούν τρισδιάστατη ανακατασκευή της εικόνας με γεωμετρική προσέγγιση στον διακριτοποιημένο (voxelized) χώρο. Η μοντελοποίηση της επίδρασης του μαγνητικού πεδίου στα εκπεμπόμενα ποζιτρόνια στοχεύει στη βελτίωση της ανάλυσης της Ποζιτρονικής Τομογραφίας και, μεταξύ άλλων, στο σχεδιασμό εξειδικευμένων πειραμάτων και μεθόδων βαθμονόμησης των συστημάτων PET. Positron range is an important spatial-resolution limiting factor in Positron Emission Tomography (PET). The effect of the magnetic field on the positron range on the PET-MRI dual modality was studied. When imaging inside a magnetic field the positron range is non-uniformly affected. Due to Lorentz Force, a decrease of the positron range is expected in the directions perpendicular to the direction of the magnetic field, whereas no variation is expected in the direction of the magnetic field. Monte Carlo simulations were performed to validate these expectations and quantify the results. GATE simulation package (version 5.0.0) was used to calculate the annihilation distribution of positrons in water, lung and rib bone. Six common positron emitters were used. The simulations were performed without and with static magnetic field set in the axial direction for various field strengths. The simulations of the positron annihilation process indicated a general reduction of the mean positron range inside the phantom volume. The evaluation of the positron annihilation distance across the directions perpendicular to the magnetic field showed a reduction of the mean positron range, whereas no increase in the positron range along the direction of the magnetic field was detected, as theoretically expected. An anaytical algorithm developed in our lab that uses a geometrical approach on the voxelized volume, allowed for the 3D reconstuction of the images derived from the GATE simulations for magnetic field strengths: 0T, 3T and 11T. The reduction of the positron range along the directions perpendicular to the magnetic field is observed only for the high energy emitters: Rb82 and Ga68. The kernels obtained from this study can be used to improve positron range correction algorithms for simultaneous PET-MRI acquisitions.

This thesis is based on the competitiveness plan of the Tampere city’s upper secondary education, which states that as a baseline, it is not a common practice to hire new employees to replace the employees who either retire or exit their employment for any other reason, and the remaining workload is aimed to be shared among the existing employees. Sharing of the tacit knowledge is emphasized in situations like this. The objective of this thesis was to clarify which facts prevent and which advance the sharing of the tacit knowledge between the teachers. The theoretical part of the thesis addresses the definition of knowledge and varieties of knowledge, know-how, sharing of tacit knowledge and the significance of the individual in sharing knowledge. How the individual behaves in sharing tacit knowledge is influenced by the individual’s personal capabilities and motivation, as well as the opportunities provided by the environment. At the individual level, the facts, which either prevent or advance sharing the tacit knowledge, are the individual’s motivation, opportunities and capability to share and receive tacit knowledge. Motivation stands for internal goals and targets, which generate the desire to share tacit knowledge. Capability stands for ability to share and receive tacit knowledge. Opportunity stands for situation specific facts, which affect sharing the tacit knowledge. From the words Motivation, Opportunities and Ability, is composed a MOA abbreviation, which is utilized as an identifier for the theory in the empirical part of this thesis. The empirical part of the thesis utilized MAO theory in identifying both the facts which advance and prevent sharing the tacit knowledge among teachers in accommodation, nutrition and household study sector. The investigation was conducted in Tampere Vocational College Tredu. Opinnäytetyö perustuu Tampereen kaupungin toisen asteen koulutuksen kilpailukykysuunnitelmaan, jossa todetaan, että eläköityvien ja muista syistä poistuvien työntekijöiden tilalle ei lähtökohtaisesti palkata uutta henkilöstöä, vaan työt pyritään jakamaan jäävien kesken. Hiljaisen tietämyksen jakaminen korostuu tällaisissa tilanteissa erityisesti. Opinnäytetyön tavoitteena oli selvittää mitkä asiat estävät ja mitkä edistävät hiljaisen tietämyksen jakamista opettajien kesken. Teoreettisessa tietopohjassa käsitellään tiedon määritelmää ja tiedon lajeja, osaamista, hiljaisen tietämyksen jakamista ja yksilön merkitystä tietämyksen jakamisessa. Yksilön käyttäytymiseen hiljaisen tietämyksen jakamisessa vaikuttavat omat henkilökohtaiset kyvyt ja motivaatiotekijät sekä ympäristön tarjoamat mahdollisuudet. Yksilötasolla hiljaisen tiedon jakamisen esteitä tai edistäviä tekijöitä ovat henkilön motivaatio, mahdollisuudet ja kyky jakaa tai ottaa vastaan hiljaista tietoa. Motivaatiolla tarkoitetaan sisäisiä päämääriä tai tavoitteita, jotka saavat aikaan halua hiljaisen tietämyksen jakamiseen. Kyvyllä tarkoitetaan taitoa hiljaisen tietämyksen jakamiseen ja vastaanottamiseen. Mahdollisuudella tarkoitetaan tilannesidonnaisia tekijöitä, jotka vaikuttavat hiljaisen tiedon jakamiseen. Sanoista Motivation (motivaatio) – Opportunity (mahdollisuus) – Ability (kyky) muodostuu MOA-lyhenne, jota käytetään tutkimuksessa käytettävän teorian tunnuksena. Tutkimuksessa selvitettiin MOA-teoriaa hyödyntäen hiljaisen tietämyksen jakamista edistäviä tai estäviä asioita Tredun majoitus- , ravitsemis- ja talousalan opettajien kesken.

El presente plan de trabajo, enmarcado en un proyecto bienal de investigación, tiene por objetivo contribuir al conocimiento sobre el tipo de prácticas áulicas que promueven y mejoran el desarrollo de la producción oral y escrita de textos. La propuesta se desarrollará mediante un diseño cuasi experimental con niños/as de primer y segundo añode primaria de una escuela pública de La Plata. En tres grados (n=90) se implementará un programa ya probado en otros contextos, que promueve el desarrollo de habilidades cognitivas, lingüísticas y socioemocionales para la alfabetización. En el grupo de comparación (n=90) de iguales características se desarrollará la materia prácticas del lenguaje que trabaja con la metodología de enseñanza de la lengua escrita del diseño curricular vigente. Ambos grupos serán evaluados en el aula con pruebas de escritura de palabras y textos pre-post intervención. Específicamente, el trabajo a cargo de la becaria consistirá en primer lugar, en la formación en aspectos teóricos ligados a la temática y al programa. Posteriormente su tarea consiste en el registro, transcripción y análisis de la implementación del programa, colaborando también en el análisis de pruebas pre-post intervención. Karakterisasi plasma nutfah mentimun dilakukan untuk mendapatkan deskripsi sifat untuk dipilih oleh pemulia sebagai bahan “koleksi kerja” yang dapat dimanfaatkan dalam program pemuliaan. Karakterisasi dilakukan terhadap 23 aksesi plasma nutfah mentimun yang ditanam di Kebun Percobaan Subang (100 m dpl) pada MK 2003, masing-masing aksesi ditanam 20 biji per petak dengan jarak tanam 70 x 40 cm. Deskripsi plasma nutfah mentimun tersebut dibuat untuk dimanfaatkan dalam upaya mendapatkan kultivar unggul baru. Dari hasil karakterisasi terpilih enam aksesi mentimun sebagai koleksi kerja pemulia berdasarkan kualitas buah dan ketahanan terhadap cekaman lingkungan. AbstactCaharcterization of Cucumis sativus germplasms was done to get the description of plant character for the breeder to use as “Working Collection” materials which were useful in breeding programme. Characterization was done on 23 accessions of Cucumis sativus germplasm planted at Subang station (100 m asl) during dry season 2003. Twenty seeds of each accession planted in a plot with planting distance of 70 x 40 cm. The description of Cucumis sativus germplasms had been done. Six accessions of Cucumis sativus were selected for Working Collection based on fruits quality and resistance to environmental stresser.

金沢大学医薬保健研究域医学系 Y染色体特異的マイナー組織適合抗原であるH-Yペプチドは男性にのみ発現し,HLA-A2に表出される.まず,HLA一致姉からの同種骨髄移植後に慢性骨髄性白血病リンパ性急性転化を来した男性患者(HLA-A2陽性)を対象に,患者の白血病細胞に対し高い細胞傷害活性を示すドナー由来のT細胞を作成した.H-Yペプチドでパルスしたドナー由来樹状細胞を抗原提示細胞に用いてドナーのT細胞を繰り返し刺激することにより,細胞傷害性T細胞を樹立した.この細胞傷害性T細胞は,ドナーの樹状細胞に対しパルスするペプチド濃度依存性の細胞傷害活性を示した.さらに,患者白血病細胞・線維芽細胞,男性リンパ芽球性リンパ細胞株細胞に対し,HLA-A2拘束性の細胞傷害活性を示した.次に,同じ男性患者が再移植を受けたのち分子遺伝学的寛解を維持していた時期の流血中に,インターフェロン(IFN)γ発現とT細胞レセプターを指標としてH-Yペプチド特異的T細胞を検出した.患者血液からCD8陽性細胞を分離し,H-Yペプチドで5時間刺激したところ,6.8%がIFN-γ陽性となった.このCD8陽性IFN-γ陽性細胞のT細胞レセプターβ鎖のCDR3領域をPCRにより増幅し,サイズ・スペクトラタイピングを行った.その結果,BV22においてin vitroで誘導された細胞傷害性T細胞と同じサイズのピークが検出された.塩基配列を決定したところ,患者の流血中に存在しているH-Yペプチド反応性T細胞とin vitroで樹立した細胞傷害性T細胞で共通のCDR3領域の配列が証明された.流血中に存在していたCD8陽性細胞は,H-Yペプチド特異的にIFN-γを発現することから,同種免疫による抗腫瘍効果の担当細胞であった可能性がある. 研究課題/領域番号:13770578, 研究期間(年度):2001-2002 出典:「INFγ発現とT細胞レセプターを指標としたマイナー組織適合抗原特異的T細胞の検出」研究成果報告書 課題番号13770578(KAKEN:科学研究費助成事業データベース(国立情報学研究所)) ( https://kaken.nii.ac.jp/ja/grant/KAKENHI-PROJECT-13770578/ )を加工して作成

Η εμβέλεια του ποζιτρονίου είναι ένας από τους κυριότερους παράγοντες περιορισμού της ανάλυσης της εικόνας στην Ποζιτρονική Τομογραφία (PET). Στην παρούσα ερευνητική εργασία μελετήθηκε η επίδραση του μαγνητικού πεδίου στην εμβέλεια του ποζιτρονίου σε διπλό σύστημα Ποζιτρονικής Τομογραφίας Εκπομπής και Μαγνητικού Τομογράφου (PET-MR) μέσω προσομοίωσης. Το ισχυρό μαγνητικό πεδίο του Μαγνητικού Τομογράφου δημιουργεί συστηματικά επηρεασμένα αποτελέσματα τα οποία διαφοροποιούνται από τα αντίστοιχα εκτός μαγνητικού πεδίου. Λόγω εμφάνισης δυνάμεων Lorentz στο εκπεμπόμενο ποζιτρόνιο αναμένεται μείωση της εμβέλειας του ποζιτρονίου στο κάθετο στο μαγνητικό πεδίο επίπεδο. Για τον έλεγχο της υπόθεσης αυτής αλλά και την ποσοτικοποίηση του αποτελέσματος, πραγματοποιήθηκαν Monte Carlo προσομοιώσεις βασισμένες στο λογισμικό GEANT4/GATE σημειακής β+ ραδιενεργού πηγής εντός απλού συστήματος PET, με εισαγωγή στατικού και ομογενούς μαγνητικού πεδίου. Για την ανακατασκευή της εικόνας χρησιμοποιήθηκαν αναλυτικοί αλγόριθμοι που έχουν αναπτυχθεί στο εργαστήριό μας (REC3D), οι οποίοι πραγματοποιούν τρισδιάστατη ανακατασκευή της εικόνας με γεωμετρική προσέγγιση στον διακριτοποιημένο (voxelized) χώρο. Η μοντελοποίηση της επίδρασης του μαγνητικού πεδίου στα εκπεμπόμενα ποζιτρόνια στοχεύει στη βελτίωση της ανάλυσης της Ποζιτρονικής Τομογραφίας και, μεταξύ άλλων, στο σχεδιασμό εξειδικευμένων πειραμάτων και μεθόδων βαθμονόμησης των συστημάτων PET. Positron range is an important spatial-resolution limiting factor in Positron Emission Tomography (PET). The effect of the magnetic field on the positron range on the PET-MRI dual modality was studied. When imaging inside a magnetic field the positron range is non-uniformly affected. Due to Lorentz Force, a decrease of the positron range is expected in the directions perpendicular to the direction of the magnetic field, whereas no variation is expected in the direction of the magnetic field. Monte Carlo simulations were performed to validate these expectations and quantify the results. GATE simulation package (version 5.0.0) was used to calculate the annihilation distribution of positrons in water, lung and rib bone. Six common positron emitters were used. The simulations were performed without and with static magnetic field set in the axial direction for various field strengths. The simulations of the positron annihilation process indicated a general reduction of the mean positron range inside the phantom volume. The evaluation of the positron annihilation distance across the directions perpendicular to the magnetic field showed a reduction of the mean positron range, whereas no increase in the positron range along the direction of the magnetic field was detected, as theoretically expected. An anaytical algorithm developed in our lab that uses a geometrical approach on the voxelized volume, allowed for the 3D reconstuction of the images derived from the GATE simulations for magnetic field strengths: 0T, 3T and 11T. The reduction of the positron range along the directions perpendicular to the magnetic field is observed only for the high energy emitters: Rb82 and Ga68. The kernels obtained from this study can be used to improve positron range correction algorithms for simultaneous PET-MRI acquisitions.

This thesis is based on the competitiveness plan of the Tampere city’s upper secondary education, which states that as a baseline, it is not a common practice to hire new employees to replace the employees who either retire or exit their employment for any other reason, and the remaining workload is aimed to be shared among the existing employees. Sharing of the tacit knowledge is emphasized in situations like this. The objective of this thesis was to clarify which facts prevent and which advance the sharing of the tacit knowledge between the teachers. The theoretical part of the thesis addresses the definition of knowledge and varieties of knowledge, know-how, sharing of tacit knowledge and the significance of the individual in sharing knowledge. How the individual behaves in sharing tacit knowledge is influenced by the individual’s personal capabilities and motivation, as well as the opportunities provided by the environment. At the individual level, the facts, which either prevent or advance sharing the tacit knowledge, are the individual’s motivation, opportunities and capability to share and receive tacit knowledge. Motivation stands for internal goals and targets, which generate the desire to share tacit knowledge. Capability stands for ability to share and receive tacit knowledge. Opportunity stands for situation specific facts, which affect sharing the tacit knowledge. From the words Motivation, Opportunities and Ability, is composed a MOA abbreviation, which is utilized as an identifier for the theory in the empirical part of this thesis. The empirical part of the thesis utilized MAO theory in identifying both the facts which advance and prevent sharing the tacit knowledge among teachers in accommodation, nutrition and household study sector. The investigation was conducted in Tampere Vocational College Tredu. Opinnäytetyö perustuu Tampereen kaupungin toisen asteen koulutuksen kilpailukykysuunnitelmaan, jossa todetaan, että eläköityvien ja muista syistä poistuvien työntekijöiden tilalle ei lähtökohtaisesti palkata uutta henkilöstöä, vaan työt pyritään jakamaan jäävien kesken. Hiljaisen tietämyksen jakaminen korostuu tällaisissa tilanteissa erityisesti. Opinnäytetyön tavoitteena oli selvittää mitkä asiat estävät ja mitkä edistävät hiljaisen tietämyksen jakamista opettajien kesken. Teoreettisessa tietopohjassa käsitellään tiedon määritelmää ja tiedon lajeja, osaamista, hiljaisen tietämyksen jakamista ja yksilön merkitystä tietämyksen jakamisessa. Yksilön käyttäytymiseen hiljaisen tietämyksen jakamisessa vaikuttavat omat henkilökohtaiset kyvyt ja motivaatiotekijät sekä ympäristön tarjoamat mahdollisuudet. Yksilötasolla hiljaisen tiedon jakamisen esteitä tai edistäviä tekijöitä ovat henkilön motivaatio, mahdollisuudet ja kyky jakaa tai ottaa vastaan hiljaista tietoa. Motivaatiolla tarkoitetaan sisäisiä päämääriä tai tavoitteita, jotka saavat aikaan halua hiljaisen tietämyksen jakamiseen. Kyvyllä tarkoitetaan taitoa hiljaisen tietämyksen jakamiseen ja vastaanottamiseen. Mahdollisuudella tarkoitetaan tilannesidonnaisia tekijöitä, jotka vaikuttavat hiljaisen tiedon jakamiseen. Sanoista Motivation (motivaatio) – Opportunity (mahdollisuus) – Ability (kyky) muodostuu MOA-lyhenne, jota käytetään tutkimuksessa käytettävän teorian tunnuksena. Tutkimuksessa selvitettiin MOA-teoriaa hyödyntäen hiljaisen tietämyksen jakamista edistäviä tai estäviä asioita Tredun majoitus- , ravitsemis- ja talousalan opettajien kesken.

El presente plan de trabajo, enmarcado en un proyecto bienal de investigación, tiene por objetivo contribuir al conocimiento sobre el tipo de prácticas áulicas que promueven y mejoran el desarrollo de la producción oral y escrita de textos. La propuesta se desarrollará mediante un diseño cuasi experimental con niños/as de primer y segundo añode primaria de una escuela pública de La Plata. En tres grados (n=90) se implementará un programa ya probado en otros contextos, que promueve el desarrollo de habilidades cognitivas, lingüísticas y socioemocionales para la alfabetización. En el grupo de comparación (n=90) de iguales características se desarrollará la materia prácticas del lenguaje que trabaja con la metodología de enseñanza de la lengua escrita del diseño curricular vigente. Ambos grupos serán evaluados en el aula con pruebas de escritura de palabras y textos pre-post intervención. Específicamente, el trabajo a cargo de la becaria consistirá en primer lugar, en la formación en aspectos teóricos ligados a la temática y al programa. Posteriormente su tarea consiste en el registro, transcripción y análisis de la implementación del programa, colaborando también en el análisis de pruebas pre-post intervención. Karakterisasi plasma nutfah mentimun dilakukan untuk mendapatkan deskripsi sifat untuk dipilih oleh pemulia sebagai bahan “koleksi kerja” yang dapat dimanfaatkan dalam program pemuliaan. Karakterisasi dilakukan terhadap 23 aksesi plasma nutfah mentimun yang ditanam di Kebun Percobaan Subang (100 m dpl) pada MK 2003, masing-masing aksesi ditanam 20 biji per petak dengan jarak tanam 70 x 40 cm. Deskripsi plasma nutfah mentimun tersebut dibuat untuk dimanfaatkan dalam upaya mendapatkan kultivar unggul baru. Dari hasil karakterisasi terpilih enam aksesi mentimun sebagai koleksi kerja pemulia berdasarkan kualitas buah dan ketahanan terhadap cekaman lingkungan. AbstactCaharcterization of Cucumis sativus germplasms was done to get the description of plant character for the breeder to use as “Working Collection” materials which were useful in breeding programme. Characterization was done on 23 accessions of Cucumis sativus germplasm planted at Subang station (100 m asl) during dry season 2003. Twenty seeds of each accession planted in a plot with planting distance of 70 x 40 cm. The description of Cucumis sativus germplasms had been done. Six accessions of Cucumis sativus were selected for Working Collection based on fruits quality and resistance to environmental stresser.
