The WATNEEDS model is based on vertical soil water balance, and considers irrigated crop area, crop calendar, and crop coefficient. In addition, the reference evaporation estimates for model inputs are based on Penman-Monteith method. In this analysis, we used climate parameters for the 2000-2010 period, and crop harvested area was kept constant using values from the MIRCA2000 dataset. Crop coefficients, growing stages and other crop parameters were based on Siebert et al. [2010].

Floodplain Ecological Response Model (FERM) is a generic conceptual model which uses completely differentiable functions to represent the continuous response of ecological targets to wetting and drying on the floodplain. It simulates a smooth response curve that avoids discontinuities (particularly in the derivative) and more closely imitates natural processes. The functions are developed based upon hypothesis and existing knowledge of response to flow conditions, sometimes in the form of a preference curve (streamflow-dependent temporal pathways of recovery or degredation). It can be coupled with a flood inundation model (taking flood inundation extents as input) to provide quantitative ecological responses. This project contains the main program that calculates the condition scores from 0 (worst) to 1 (best) to represent the healthiness of the vegetation. The sample data folder contains a complete test including input data, parameters and output data. The input dry-wet-spell sequencing .tif files are output from the calculate_stats function in the ferm_main.py. B16F10/OVA H2B-mCherry cells are intradermally injected into the back skin of mice. To assess the impact of the immunomodulatory stroma on immunotherapy at different tumor regions (core, rim, invasion strands), adoptive T cell transfer (OT-I dsRed cytotoxic T cells) is given. At endpoint, tumors are microdissected into core and rim + invasion strands separately and digested to single-cell suspensions in preparation for RNA sequencing. Here we show data corresponding to the rim + invasion strands. B16F10/OVA H2B-mCherry mouse melanoma tumor cells and OT-I dsRed cytotoxic T cells can be sorted based on their fluorescence in 384 well plates for single cell SORTseq. Unstained and viability stained only single cell suspensions of mouse skin are used as controls for the viability staining. The cells of interest are initially gated by size and fluorescence-based on pure in-vitro cell cultures (either single-cell suspensions of a single cell line or single-cell suspensions comprised of both B16F10/OVA H2B-mCherry and OT-I dsRed cytotoxic T cells). Size-based gates were further applied to the samples consisting of single-cell suspensions of digested tumor tissue. In those gates further gating to eliminate doublets and dead cells was applied. Eventually, only cells positive for either H2B-mCherry (B16F10/OVA melanoma tumor cells) or dsRed (OT-I cytotoxic T cells) are sorted out in a single-cell manner (one cell per well).

The original ICT Skill Test application is a test instrument for evaluating digital skills. The implemented test application is a web application, written in PHP and JavaScript programming languages together with TinyMVC- and Bootstrap-frameworks. The software runs on a system with apache, php 5.6 and postgreql 9.2 or newer database. Apache needs php and rewrite modules. The test includes a survey phase, which collects background information, and 17 test items categorised into the following categories: basic digital skills (word processing, spreadsheet, social networking, information seeking, presentation, basic use of computers, image processing, and web content creation), advanced technical skills (operating system installation and initialisation, maintenance and updating, software installation and initialisation, information security, and information networks) and professional ICT skills (server environments, database operations, digital technology, and programming). The test content is in Finnish only. List of configuration files which needs to be configured for running php application: * myapp/configs/application.php -This file includes site, language, security and ldap configurations. * myapp/configs/database.php -This file includes database configurations. * htdock/.htaccess -This file has base url rewrite rule. Scrip for database creation is located in the db directory.
