This rapid mixing underlies Landau damping of these fluctuations in a quiescent plasma—one of the most fundamental physical phenomena that makes plasma different from a conventional fluid. Nevertheless, broad power law spectra of compressive fluctuations are observed in turbulent astrophysical plasmas (most vividly, in the solar wind) under conditions conducive to strong Landau damping. Elsewhere in nature, such spectra are normally associated with fluid turbulence, where energy cannot be dissipated in the inertial-scale range and is, therefore, cascaded from large scales to small. By direct numerical simulations and theoretical arguments, it is shown here that turbulence of compressive fluctuations in collisionless plasmas strongly resembles one in a collisional fluid and does have broad power law spectra. This “fluidization” of collisionless plasmas occurs, because phase mixing is strongly suppressed on average by “stochastic echoes,” arising due to nonlinear advection of the particle distribution by turbulent motions. Other than resolving the long-standing puzzle of observed compressive fluctuations in the solar wind, our results suggest a conceptual shift for understanding kinetic plasma turbulence generally: rather than being a system where Landau damping plays the role of dissipation, a collisionless plasma is effectively dissipationless, except at very small scales. The universality of “fluid” turbulence physics is thus reaffirmed even for a kinetic, collisionless system.

Distribution of entanglement over long distances can be enabled through coherently interfacing qubit pairs via photonic channels. Here, we report the realization of optically generated quantum entanglement between electron spin qubits confined in two distant semiconductor quantum dots. The protocol relies on spin-photon entanglement in the trionic Λ system and quantum erasure of the Raman-photon path information. The measurement of a single Raman photon is used to project the spin qubits into a joint quantum state with an interferometrically stabilized and tunable relative phase. We report an average Bell-state fidelity for |ψ^{(+)}⟩ and |ψ^{(-)}⟩ states of 61.6±2.3% and a record-high entanglement generation rate of 7.3 kHz between distant qubits. It fully accounts for the Coulomb distortions of electron trajectories, does not require sculpted fields to retrieve phase information and, in principle, is applicable to a broad range of electron momenta. By comparatively measuring the differential photoelectron spectra from strong-field ionization of ${\mathrm{N}}_{2}$ molecules and their companion atoms of Ar, some photoelectron holography patterns are found to be dephased for both targets. This is well reproduced by the full-dimensional time-dependent Schr\"odinger equation and the Coulomb quantum-orbit strong-field approximation (CQSFA) simulation. Using the CQSFA, we trace back our observations to different parities of the $3p$ orbital of Ar and the highest-occupied molecular orbital of ${\mathrm{N}}_{2}$ via interfering Coulomb-distorted quantum orbits carrying different initial phases. This method could in principle be used to extract bound-state phases from any holographic structure, with a wide range of potential applications in recollision physics and spectroscopy.

Wilson; Adolfo Saiardi;Inositol phosphates are a large and diverse family of signalling molecules. While genetic studies have discovered important functions for them, the biochemistry behind these roles is often not fully characterized. A key obstacle in inositol phosphate research in mammalian cells has been the lack of straightforward techniques for their purification and analysis. Here we describe the ability of titanium dioxide (TiO2) beads to bind inositol phosphates. This discovery allowed the development of a new purification protocol that, coupled with gel analysis, permitted easy identification and quantification of InsP6 (phytate), its pyrophosphate derivatives InsP7 and InsP8, and the nucleotides ATP and GTP from cell or tissue extracts. Using this approach, InsP6, InsP7 and InsP8 were visualized in Dictyostelium extracts and a variety of mammalian cell lines and tissues, and the effects of metabolic perturbation on these were explored. TiO2 bead purification also enabled us to quantify InsP6 in human plasma and urine, which led to two distinct but related observations. Firstly, there is an active InsP6 phosphatase in human plasma, and secondly, InsP6 is undetectable in either fluid. These observations seriously question reports that InsP6 is present in human biofluids and the advisability of using InsP6 as a dietary supplement.

Green tea is consumed globally and is reported to have anti-inflammatory properties, which may be mediated through the impact on cyclooxygenase and lipoxygenase pathways. Recent data suggest that green tea catechins (GTCs) reduce acute UVR effects, but human trials examining their photoprotective potential are scarce. Objective: We performed a double-blind, randomized, placebo-controlled trial to examine whether GTCs protect against clinical, histologic, and biochemical indicators of UVR-induced inflammation. Design: Healthy adults (aged 18–65 y, phototypes I–II) were randomly allocated to 1350 mg encapsulated green tea extract (540 mg GTC) with 50 mg vitamin C or placebo twice daily for 3 mo. Impact on skin erythema, dermal leukocytic infiltration, and concentrations of proinflammatory eicosanoids was assessed after solar-simulated UVR challenge, and subject compliance was determined through assay of urinary GTC metabolite epigallocatechin glucuronide. Results: Volunteers were assigned to the active (n = 25) or the placebo (n = 25) group. After supplementation, median (IQR) sunburn threshold (minimal erythema dose) was 28 (20–28) and 20 (20–28) mJ/cm2 in the active and placebo groups, respectively (nonsignificant), with no difference in AUC analysis for measured erythema index after a geometric series of 10 UVR doses. Skin immunohistochemistry showed increased neutrophil and CD3+ T-lymphocyte numbers post-UVR in both groups (P < 0.01) with no statistically significant differences between groups after supplementation. Cyclooxygenase and lipoxygenase metabolites prostaglandin E2 (vasodilator) and 12-hydroxyeicosatetraenoicacid (chemoattractant), respectively, increased after UVR (P < 0.05), with no differences between supplementation groups. Conclusion: Oral GTC (1080 mg/d) with vitamin C over 3 mo did not significantly reduce skin erythema, leukocyte infiltration, or eicosanoid response to UVR inflammatory challenge. This study investigates the emotional content of 174 posts from 162 posters to online forums made by young (age 14–21) first‐time mothers to understand what emotions are expressed in these posts and how these emotions interact with the types of posts and the indicators of Information Poverty within the posts. Using textual analyses we provide a classification of emotions within posts across three main themes of interaction emotions, response emotions, and preoccupation emotions and show that many requests for information by young first‐time mothers are motivated by negative emotions. This has implications for how moderators of online news groups respond to online request for help and for understanding how to support vulnerable young parents.

The suggested approach makes use of an initial line search along the hyper-diagonal of the design space in order to find a suitable starting point for a subsequent gradient based optimisation algorithm. During the optimisation an upper bound constraint is imposed on the condition number of the correlation matrix in order to keep it from being ill conditioned. Partial derivatives of both the condensed log likelihood function and the condition number are obtained using the adjoint method, the latter has been derived in this work. The approach is tested on a number of analytical examples and comparisons are made to other optimisation approaches. Finally the approach is used to construct metamodels for a finite element model of an aircraft wing box comprising of 126 thickness design variables and is then compared with a sub-set of the other optimisation approaches.

Millen;eLife digest CHARGE syndrome is a rare genetic condition that causes various developmental abnormalities, including heart defects, deafness and neurological defects. In most cases, it is caused by mutations in a human gene called CHD7. CHD7 is known to control the expression of other genes during embryonic development, but the molecular mechanisms by which mutations in CHD7 lead to the neural defects found in CHARGE syndrome are unclear. During embryonic development, the neural tube—the precursor to the nervous system—is divided into segments, which give rise to different neural structures. The r1 segment, for example, forms the cerebellum, and the secretion of a protein called FGF8 (short for fibroblast growth factor 8) by a nearby structure called the isthmus organiser has an important role in this process. Since a reduction in FGF8 causes defects similar to those found in CHARGE syndrome, Yu et al. decided to investigate if the FGF signalling pathway was involved in this syndrome. Mice should have two working copies of the Chd7 gene, and mice that lack one of these suffer from symptoms similar to those of humans with CHARGE syndrome. Yu et al. examined the embryos of these mice and found that the isthmus organiser produced less FGF8. Embryos with no working copies of the gene completely lost the r1 segment. The loss of this segment appeared to be caused by changes in the expression of homeobox genes (the genes that determine the identity of brain segments). Embryos that did not have any working copies of the Chd7 gene died early in development, which made further studies impossible. However, embryos that had one working copy of the Chd7 gene survived, and Yu et al. took advantage of this to study the effects of reduced FGF8 expression on these mice. These experiments showed that mice with just one working copy of the Fgf8 gene and one working copy of the Chd7 gene had a small cerebellar vermis. This part of the cerebellum is known to be very sensitive to changes in FGF8 signalling. Yu et al. then used an MRI scanner to look at the cerebellar vermis in patients with CHARGE syndrome, and found that more than half of the patients had abnormal cerebella. In addition to confirming that studies on mouse embryos can provide insights into human disease, the work of Yu et al. add defects in the cerebellar vermis to the list of developmental abnormalities associated with CHARGE syndrome. The next step will be to test if any mutations in the human FGF8 gene can contribute to cerebellar defects in CHARGE syndrome, and to investigate if any other developmental defects in CHARGE syndrome are associated with abnormal FGF8 levels. DOI: http://dx.doi.org/10.7554/eLife.01305.002 Mutations in CHD7 are the major cause of CHARGE syndrome, an autosomal dominant disorder with an estimated prevalence of 1/15,000. We have little understanding of the disruptions in the developmental programme that underpin brain defects associated with this syndrome. Using mouse models, we show that Chd7 haploinsufficiency results in reduced Fgf8 expression in the isthmus organiser (IsO), an embryonic signalling centre that directs early cerebellar development. Consistent with this observation, Chd7 and Fgf8 loss-of-function alleles interact during cerebellar development. CHD7 associates with Otx2 and Gbx2 regulatory elements and altered expression of these homeobox genes implicates CHD7 in the maintenance of cerebellar identity during embryogenesis. Finally, we report cerebellar vermis hypoplasia in 35% of CHARGE syndrome patients with a proven CHD7 mutation. These observations provide key insights into the molecular aetiology of cerebellar defects in CHARGE syndrome and link reduced FGF signalling to cerebellar vermis hypoplasia in a human syndrome. Although the effects of light on brain network activity in humans are well demonstrated, the effects of light on cognitive performance are inconclusive, with the size, as well as direction, of the effect depending on the nature of the task. Similarly, in nocturnal rodents, bright light can either facilitate or disrupt performance depending on the type of task employed. Crucially, it is unclear whether the effects of light on behavioural performance are mediated via the classical image-forming rods and cones or the melanopsin-expressing photosensitive retinal ganglion cells. Here, we investigate the modulatory effects of light on memory performance in mice using the non-aversive, spontaneous recognition task. Importantly, we examine which photoreceptors are required to mediate the effects of light on memory performance. By using a cross-over design, we show that object recognition memory is disrupted when the test phase is conducted under a bright light (350 lux), regardless of the light level in the sample phase (10 or 350 lux), demonstrating that exposure to a bright light at the time of test, rather than at the time of encoding, impairs performance. Strikingly, the modulatory effect of light on memory performance is completely abolished in both melanopsin-deficient and rodless–coneless mice. Our findings provide direct evidence that melanopsin-driven and rod/cone-driven photoresponses are integrated in order to mediate the effect of light on memory performance.

This rapid mixing underlies Landau damping of these fluctuations in a quiescent plasma—one of the most fundamental physical phenomena that makes plasma different from a conventional fluid. Nevertheless, broad power law spectra of compressive fluctuations are observed in turbulent astrophysical plasmas (most vividly, in the solar wind) under conditions conducive to strong Landau damping. Elsewhere in nature, such spectra are normally associated with fluid turbulence, where energy cannot be dissipated in the inertial-scale range and is, therefore, cascaded from large scales to small. By direct numerical simulations and theoretical arguments, it is shown here that turbulence of compressive fluctuations in collisionless plasmas strongly resembles one in a collisional fluid and does have broad power law spectra. This “fluidization” of collisionless plasmas occurs, because phase mixing is strongly suppressed on average by “stochastic echoes,” arising due to nonlinear advection of the particle distribution by turbulent motions. Other than resolving the long-standing puzzle of observed compressive fluctuations in the solar wind, our results suggest a conceptual shift for understanding kinetic plasma turbulence generally: rather than being a system where Landau damping plays the role of dissipation, a collisionless plasma is effectively dissipationless, except at very small scales. The universality of “fluid” turbulence physics is thus reaffirmed even for a kinetic, collisionless system.
