    ABSTRACT The cell shape of Trypanosoma brucei is influenced by flagellum-to-cell-body attachment through a specialised structure – the flagellum attachment zone (FAZ). T. brucei exhibits numerous morphological forms during its life cycle and, at each stage, the FAZ length varies. We have analysed FLAM3, a large protein that localises to the FAZ region within the old and new flagellum. Ablation of FLAM3 expression causes a reduction in FAZ length; however, this has remarkably different consequences in the tsetse procyclic form versus the mammalian bloodstream form. In procyclic form cells FLAM3 RNAi results in the transition to an epimastigote-like shape, whereas in bloodstream form cells a severe cytokinesis defect associated with flagellum detachment is observed. Moreover, we demonstrate that the amount of FLAM3 and its localisation is dependent on ClpGM6 expression and vice versa. This evidence demonstrates that FAZ is a key regulator of trypanosome shape, with experimental perturbations being life cycle form dependent. An evolutionary cell biology explanation suggests that these differences are a reflection of the division process, the cytoskeleton and intrinsic structural plasticity of particular life cycle forms. Summary: Trypanosoma brucei FLAM3 is a flagellar FAZ protein. Its depletion leads to a reduction in FAZ length, which has different consequences depending on the life cycle stage of the parasite.

    In this paper, the problem of joint design of Spectrum Sensing (SS) and receive beamforming (BF), with reference to a Cognitive Radio (CR) system, is considered. The aim of the proposed design is the maximization of the achievable average uplink rate of a Secondary User (SU), subject to an outage-based Quality-of-Service (QoS) constraint for primary communication. A hybrid CR system approach is studied, according to which, the system either operates as an interweave (i.e., opportunistic) or as an underlay (i.e., spectrum sharing) CR system, based on SS results. A realistic Channel State Information (CSI) framework is assumed, according to which, the direct channel links are known by the multiple antenna receivers (RXs), while, merely statistical (covariance) information is available for the interference links. A new, closed form approximation is derived for the outage probability of primary communication, and the problem of rate-optimal selection of SS parameters and receive beamformers is addressed for hybrid, interweave and underlay CR systems. It is proven that our proposed system design outperforms both underlay and interweave CR systems for a range of system scenarios.

    Summarization: This paper presents the design procedure of a wideband UHF RFID reader antenna with polarization diversity suitable for searching tagged items. The antenna consists of a microstrip array with alternating orthogonal bow-tie elements, which are fed in series by a pair of microstrip lines. The elements are designed to provide the required bandwidth. The inter-element distance is adjusted to be about 0.8λ at the center frequency, whereas the elements provide in-phase excitation and create two orthogonal electric field components that give beams with polarization diversity. Simulated results indicate that the bandwidth covers the frequency bands of ETSI and FCC (865-928 MHz) standards. Regarding the coverage, it was deduced that the antenna can identify tagged items in a semi-cylindrical volume around its axis. The radius of coverage depends on the input power and tag sensitivity.     Background\ud The value of the nosological distinction between non-affective and affective psychosis has frequently been challenged. We aimed to investigate the transdiagnostic dimensional structure and associated characteristics of psychopathology at First Episode Psychosis (FEP). Regardless of diagnostic categories, we expected that positive symptoms occurred more frequently in ethnic minority groups and in more densely populated environments, and that negative symptoms were associated with indices of neurodevelopmental impairment.\ud \ud Method\ud This study included 2182 FEP individuals recruited across six countries, as part of the EUropean network of national schizophrenia networks studying Gene–Environment Interactions (EU-GEI) study. Symptom ratings were analysed using multidimensional item response modelling in Mplus to estimate five theory-based models of psychosis. We used multiple regression models to examine demographic and context factors associated with symptom dimensions.\ud \ud Results\ud A bifactor model, composed of one general factor and five specific dimensions of positive, negative, disorganization, manic and depressive symptoms, best-represented associations among ratings of psychotic symptoms. Positive symptoms were more common in ethnic minority groups. Urbanicity was associated with a higher score on the general factor. Men presented with more negative and less depressive symptoms than women. Early age-at-first-contact with psychiatric services was associated with higher scores on negative, disorganized, and manic symptom dimensions.\ud \ud Conclusions\ud Our results suggest that the bifactor model of psychopathology holds across diagnostic categories of non-affective and affective psychosis at FEP, and demographic and context determinants map onto general and specific symptom dimensions. These findings have implications for tailoring symptom-specific treatments and inform research into the mood-psychosis spectrum.

    Summarization: Fused Deposition Modelling (FDM) is a simple additive manufacturing (AM) technology utilising fine filament extrusion of mainly thermoplastic materials to build 3D objects layer by layer. The focus of this paper is firstly to design hierarchical honeycombs and secondly to fabricate such structures using FDM with polylactic acid (PLA). The manufactured hierarchical honeycombs were tested under compression. The compression performance of the hierarchical honeycombs was assessed also with finite element analysis and the results were compared with the experimental derived properties in order to elucidate the mechanical behaviour of such honeycomb AM structures. The results reveal that for the 2nd order hierarchy of honeycombs the stiffness and the strength are increased in comparison with the 0th and the 1st hierarchies.     Summarization: Alkali activated materials (AAMs), also named geopolymers or inorganic polymers, are materials that are produced when alkaline solutions react with precursors containing aluminosilicate phases. The most commonly used alkaline solutions, include sodium or potassium hydroxides and/or silicates, while aluminosilicates may include suitable raw materials and by-products/wastes produced from various industrial processes. AAMs can be commercialized in various industrial sectors. When AAMs are produced with the use of industrial wastes, including for example metallurgical slags of various types, fly ashes, bauxite residues, construction and demolition wastes or leaching residues, the benefits are twofold. They also include the valorization of these wastes for the production of value added products and the substantial reduction of the environmental footprint of the construction sector, due to the considerable savings of virgin raw materials. This Special Issue of Minerals includes 14 papers highlighting the advances and future trends in alkali activation of wastes for the production of AAMs that can be used as sensors, adsorbents, catalysts, pH modifiers, as well as for the production of zeolite foams, low carbon binders, cemented paste backfill, alkali activated mortars and concrete. Furthermore, the elucidation of mechanisms involved during alkali activation, as well as the mineralogy, microstructure and properties of AAMs with the use of advanced techniques are discussed. All these papers aim to attract the interest of the readers and especially of young scientists and students at graduate and post-graduate levels in this very important scientific field.     Summarization: Accurate and reliable flow forecasting in complex Canadian prairie watersheds has been one of the major challenges faced by hydrologists. In an attempt to improve the accuracy and reliability of a reservoir inflow forecast, this study investigates structurally different hydrological models along with ensemble precipitation forecasts to identify the most skillful and reliable model. The key goal is to assess whether short- and medium-range ensemble flood forecasting in large complex basins can be accurately achieved by simple conceptual lumped models (e.g., SACSMA with SNOW17 and MACHBV with SNOW17) or it requires a medium level distributed model (e.g., WATFLOOD) or an advanced macroscale land-surface based model (VIC coupled with routing module (RVIC)). Eleven (11)-member precipitation forecasts from second-generation Global Ensemble Forecast System reforecast (GEFSv2) were used as inputs. Each of the ensemble members was bias-corrected by Empirical Quantile Mapping method using the Canadian Precipitation Analysis (CaPA) as a training/verification dataset. Forecast evaluation is performed for 1-day up to 8-days forecast lead times in a 6-month hindcast period. Results indicate that bias-correcting precipitation forecasts using verifying datasets (such as CaPA) for a training period of at least two years before the forecast time, produces skillful ensemble hydrological forecasts. A comparison of models in forecast mode shows that the two lumped models (SACSMA and MACHBV) can provide better overall forecast performance than the benchmark WATFLOOD and the macroscale Variable Infiltration Capacity (VIC) model. However, for shorter lead-times, particularly up to day 3, the benchmark distributed model provides competitive reliability, as compared to the lumped models.     ABSTRACT The cell shape of Trypanosoma brucei is influenced by flagellum-to-cell-body attachment through a specialised structure – the flagellum attachment zone (FAZ). T. brucei exhibits numerous morphological forms during its life cycle and, at each stage, the FAZ length varies. We have analysed FLAM3, a large protein that localises to the FAZ region within the old and new flagellum. Ablation of FLAM3 expression causes a reduction in FAZ length; however, this has remarkably different consequences in the tsetse procyclic form versus the mammalian bloodstream form. In procyclic form cells FLAM3 RNAi results in the transition to an epimastigote-like shape, whereas in bloodstream form cells a severe cytokinesis defect associated with flagellum detachment is observed. Moreover, we demonstrate that the amount of FLAM3 and its localisation is dependent on ClpGM6 expression and vice versa. This evidence demonstrates that FAZ is a key regulator of trypanosome shape, with experimental perturbations being life cycle form dependent. An evolutionary cell biology explanation suggests that these differences are a reflection of the division process, the cytoskeleton and intrinsic structural plasticity of particular life cycle forms. Summary: Trypanosoma brucei FLAM3 is a flagellar FAZ protein. Its depletion leads to a reduction in FAZ length, which has different consequences depending on the life cycle stage of the parasite.

    In this paper, the problem of joint design of Spectrum Sensing (SS) and receive beamforming (BF), with reference to a Cognitive Radio (CR) system, is considered. The aim of the proposed design is the maximization of the achievable average uplink rate of a Secondary User (SU), subject to an outage-based Quality-of-Service (QoS) constraint for primary communication. A hybrid CR system approach is studied, according to which, the system either operates as an interweave (i.e., opportunistic) or as an underlay (i.e., spectrum sharing) CR system, based on SS results. A realistic Channel State Information (CSI) framework is assumed, according to which, the direct channel links are known by the multiple antenna receivers (RXs), while, merely statistical (covariance) information is available for the interference links. A new, closed form approximation is derived for the outage probability of primary communication, and the problem of rate-optimal selection of SS parameters and receive beamformers is addressed for hybrid, interweave and underlay CR systems. It is proven that our proposed system design outperforms both underlay and interweave CR systems for a range of system scenarios.

    Summarization: This paper presents the design procedure of a wideband UHF RFID reader antenna with polarization diversity suitable for searching tagged items. The antenna consists of a microstrip array with alternating orthogonal bow-tie elements, which are fed in series by a pair of microstrip lines. The elements are designed to provide the required bandwidth. The inter-element distance is adjusted to be about 0.8λ at the center frequency, whereas the elements provide in-phase excitation and create two orthogonal electric field components that give beams with polarization diversity. Simulated results indicate that the bandwidth covers the frequency bands of ETSI and FCC (865-928 MHz) standards. Regarding the coverage, it was deduced that the antenna can identify tagged items in a semi-cylindrical volume around its axis. The radius of coverage depends on the input power and tag sensitivity.     Background\ud The value of the nosological distinction between non-affective and affective psychosis has frequently been challenged. We aimed to investigate the transdiagnostic dimensional structure and associated characteristics of psychopathology at First Episode Psychosis (FEP). Regardless of diagnostic categories, we expected that positive symptoms occurred more frequently in ethnic minority groups and in more densely populated environments, and that negative symptoms were associated with indices of neurodevelopmental impairment.\ud \ud Method\ud This study included 2182 FEP individuals recruited across six countries, as part of the EUropean network of national schizophrenia networks studying Gene–Environment Interactions (EU-GEI) study. Symptom ratings were analysed using multidimensional item response modelling in Mplus to estimate five theory-based models of psychosis. We used multiple regression models to examine demographic and context factors associated with symptom dimensions.\ud \ud Results\ud A bifactor model, composed of one general factor and five specific dimensions of positive, negative, disorganization, manic and depressive symptoms, best-represented associations among ratings of psychotic symptoms. Positive symptoms were more common in ethnic minority groups. Urbanicity was associated with a higher score on the general factor. Men presented with more negative and less depressive symptoms than women. Early age-at-first-contact with psychiatric services was associated with higher scores on negative, disorganized, and manic symptom dimensions.\ud \ud Conclusions\ud Our results suggest that the bifactor model of psychopathology holds across diagnostic categories of non-affective and affective psychosis at FEP, and demographic and context determinants map onto general and specific symptom dimensions. These findings have implications for tailoring symptom-specific treatments and inform research into the mood-psychosis spectrum.
