The data were computed using the Truncated Conformal Space Approach (TCSA), which is a Hamiltonian truncation method. Parameters: Cosine frequencies: β = 2.49239 for SG and β1 = 1.01066 and β2 = 2.49239 for DSG Dimensionless (mass)⨉(system size) parameter (m: first SG breather mass, L: system size): mL = 0.01, 0.1, 1, 2, 5 TCSA details: truncation basis: free massless boson CFT with Dirichlet boundary conditions, restricted to the ground state symmetry sector truncation cutoff (maximum CFT energy shell): 42 basis size: 85674 The spectra correspond to the full list of eigenvalues of the truncated Hamiltonian matrices in increasing order, and the eigenvectors correspond to matrices of dimensions 5173⨉5173, corresponding to the components of the lowest 5173 energy levels in the lowest 5173 basis states (the best convergent part of the eigenvector matrix at the top left corner). Each eigenvector corresponds to a column of the above matrices, in the same order as the eigenvalues. Depending on laser pulse energies and processing times, this allowed for different modes of surface modification. At low laser intensities, an etching almost without thermal impact was detected, whereas higher laser intensities led to the generation of hierarchical micro and nanostructures. The usage of argon or atmospheric air as cover gases during the laser processing had only minor effects on the surface structures. Observed differences in the etching or functionalization of the wooden surfaces mostly originated in the chemical structure of the surface finish and the physical properties of the wood substrates, such as the density or moisture content. Obiecującą perspektywą dla skuteczności leczenia nowotworów jest łączenie klasycznej chemioterapii z terapią ukierunkowaną na różne cele molekularne. Zastosowanie inhibitorów białek zaangażowanych w ścieżki sygnalizacyjne, związane z powstawaniem i rozwojem nowotworu, może wpłynąć na skuteczność i specyficzność stosowanych leków. Fosfataza DUSP6 jest białkiem, którego rola w procesie nowotworzenia jest niejednoznaczna. Wskazuje się zarówno na jej właściwości supresyjne, jak i onkogenne. Celem projektu badawczego była ocena wpływu inhibitora fosfatazy DUSP6 (BCI) na stopień cytotoksyczności onkoterapeutyków – irinotekanu i oksaliplatyny w komórkach raka jelita grubego, w układzie in vitro. W badaniu wykorzystano dwie ludzkie linie komórkowe, różniące się statusem genu BRCA2, a tym samym zdolnością do naprawy uszkodzeń DNA – DLD1 oraz DLD1 BRCA2 (-/-). Układy badawcze rozszerzono o zastosowanie inhibitora kinazy serynowo-treoninowej ATM (KU60019), będącej jednym z kluczowych białek mechanizmu naprawy DNA. W pierwszym etapie badań, dokonano oceny cytotoksyczności (test MTT) pojedynczych związków, a następnie ich układów. Oceniono wpływ wybranych stężeń BCI na właściwości chemioterapeutyków, również w obecności inhibitora ATM. W kolejnym etapie, wybrane układy doświadczalne poddano analizie cytometrycznej (Alexa Fluor®488 Annexin V/Dead Cell Apoptosis Kit), celem sprawdzenia ich aktywności w badanych komórkach oraz wskazania, jakiego rodzaju śmierć komórkową indukują. Combining traditional chemotherapy with treatments that focus on different molecular targets is a useful approach to enhance the effectiveness of cancer treatments. The efficiency and specificity of drugs can be affected by the use of inhibitors of proteins that are involved in signaling pathways associated with cancer development. DUSP6 is a phosphatase protein with an ambiguous role in tumorigenesis. It was found to have both suppressive and oncogenic properties. This research project aimed to evaluate the effect of the phosphatase DUSP6 inhibitor (BCI) on the cytotoxicity of the therapeutical agents used in cancer treatment (irinotecan and oxaliplatin) in colorectal cancer cells in vitro. Two cell lines differing in BRCA2 gene status and DNA repair capacity were used in the study – DLD1 and DLD1 BRCA2 (-/-). The research was extended with the use of ATM kinase inhibitor (KU60019), one of the key proteins involved in cellular DNA repair mechanisms. The cytotoxicity of the individual compounds (MTT assay) was analyzed in the first stage of the study, followed by the investigation of the cytotoxicity of the compounds in combinations. The effect of selected concentrations of BCI on the properties of chemotherapeutics was assessed, also in the presence of an ATM inhibitor. In the next step, selected compound combinations were subjected to cytometric analysis (Alexa Fluor®488 Annexin V/Dead Cell Apoptosis Kit) to evaluate their activity in the studied cells and to indicate the type of cell death that they can induce. The data were computed using the Truncated Conformal Space Approach (TCSA), which is a Hamiltonian truncation method. Parameters: Cosine frequencies: β = 2.49239 for SG and β1 = 1.01066 and β2 = 2.49239 for DSG Dimensionless (mass)⨉(system size) parameter (m: first SG breather mass, L: system size): mL = 0.01, 0.1, 1, 2, 5 TCSA details: truncation basis: free massless boson CFT with Dirichlet boundary conditions, restricted to the ground state symmetry sector truncation cutoff (maximum CFT energy shell): 42 basis size: 85674 The spectra correspond to the full list of eigenvalues of the truncated Hamiltonian matrices in increasing order, and the eigenvectors correspond to matrices of dimensions 5173⨉5173, corresponding to the components of the lowest 5173 energy levels in the lowest 5173 basis states (the best convergent part of the eigenvector matrix at the top left corner). Each eigenvector corresponds to a column of the above matrices, in the same order as the eigenvalues. Zbiór danych koincydencyjnych w postaci macierzy Egamma-Ex (energia kwantów gamma, wyrażona w keV, vs. energia wzbudzenia jądra 13C, wyrażona w MeV) zgromadzonych podczas eksperymentu rozpraszania nieelastycznego wiązki protonów na tarczy 13C („Study of M4 stretched configuration decay in 13C”) przeprowadzonego w CCB IFJ PAN. Zapis danych zgodny z formatem ROOT. The set of coincidence data in a form of the Egamma-Ex matrix (gamma energy, in keV, vs. excitation energy of the 13C nucleus, in MeV) collected during the experiment with inelastic scattering of proton beam on the 13C target („Study of M4 stretched configuration decay in 13C”) performed at CCB IFJ PAN. Depending on laser pulse energies and processing times, this allowed for different modes of surface modification. At low laser intensities, an etching almost without thermal impact was detected, whereas higher laser intensities led to the generation of hierarchical micro and nanostructures. The usage of argon or atmospheric air as cover gases during the laser processing had only minor effects on the surface structures. Observed differences in the etching or functionalization of the wooden surfaces mostly originated in the chemical structure of the surface finish and the physical properties of the wood substrates, such as the density or moisture content.
