    AbstractBackgroundBombyx mori silk fibroin is a biomacromolecule that allows the assembly of scaffolds for tissue engineering and regeneration purposes due to its cellular adhesiveness, high biocompatibility and low immunogenicity. Earlier work showed that two types of 3D silk fibroin nonwovens (3D-SFnws) implanted into mouse subcutaneous tissue were promptly vascularized via undefined molecular mechanisms. The present study used nontumorigenic adult human dermal fibroblasts (HDFs) adhering to a third type of 3D-SFnws to assess whether HDFs release exosomes whose contents promote neoangiogenesis.MethodsElectron microscopy imaging and physical tests defined the features of the novel carded/hydroentangled 3D-SFnws. HDFs were cultured on 3D-SFnws and polystyrene plates in an exosome-depleted medium. DNA amounts and D-glucose consumption revealed the growth and metabolic activities of HDFs on 3D-SFnws. CD9-expressing total exosome fractions were from conditioned media of 3D-SFnws and 2D polystyrene plates HDF cultures. Angiogenic growth factors (AGFs) in equal amounts of the two groups of exosomal proteins were analysed via double-antibody arrays. A tube formation assay using human dermal microvascular endothelial cells (HDMVECs) was used to evaluate the exosomes’ angiogenic power.ResultsThe novel features of the 3D-SFnws met the biomechanical requirements typical of human soft tissues. By experimental day 15, 3D-SFnws-adhering HDFs had increased 4.5-fold in numbers and metabolized 5.4-fold more D-glucose than at day 3 in vitro. Compared to polystyrene-stuck HDFs, exosomes from 3D-SFnws-adhering HDFs carried significantly higher amounts of AGFs, such as interleukin (IL)-1α, IL-4 and IL-8; angiopoietin-1 and angiopoietin-2; angiopoietin-1 receptor (or Tie-2); growth-regulated oncogene (GRO)-α, GRO-β and GRO-γ; matrix metalloproteinase-1; tissue inhibitor metalloproteinase-1; and urokinase-type plasminogen activator surface receptor, but lesser amounts of anti-angiogenic tissue inhibitor metalloproteinase-2 and pro-inflammatory monocyte chemoattractant protein-1. At concentrations from 0.62 to 10 μg/ml, the exosomes from 3D-SFnws-cultured HDFs proved their angiogenic power by inducing HDMVECs to form significant amounts of tubes in vitro.ConclusionsThe structural and mechanical properties of carded/hydroentangled 3D-SFnws proved their suitability for tissue engineering and regeneration applications. Consistent with our hypothesis, 3D-SFnws-adhering HDFs released exosomes carrying several AGFs that induced HDMVECs to promptly assemble vascular tubes in vitro. Hence, we posit that once implanted in vivo, the 3D-SFnws/HDFs interactions could promote the vascularization and repair of extended skin wounds due to burns or other noxious agents in human and veterinary clinical settings.

    Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive tumors; it is often diagnosed at an advanced stage and is hardly treatable. These issues are strictly linked to the absence of early diagnostic markers and the low efficacy of treatment approaches. Recently, the study of the metabolic alterations in cancer cells has opened the way to important findings that can be exploited to generate new potential therapies. Within this scenario, mitochondria represent important organelles within which many essential functions are necessary for cell survival, including some key reactions involved in energy metabolism. These organelles remodel their shape by dividing or fusing themselves in response to cellular needs or stimuli. Interestingly, many authors have shown that mitochondrial dynamic equilibrium is altered in many different tumor types. However, up to now, it is not clear whether PDAC cells preferentially take advantage of fusion or fission processes since some studies reported a wide range of different results. This review described the role of both mitochondria arrangement processes, i.e., fusion and fission events, in PDAC, showing that a preference for mitochondria fragmentation could sustain tumor needs. In addition, we also highlight the importance of considering the metabolic arrangement and mitochondria assessment of cancer stem cells, which represent the most aggressive tumor cell type that has been shown to have distinctive metabolic features to that of differentiated tumor cells.

    This paper address the problem of Stavrogin’s ambivalence and Walter Benjamin’s interpretation of Dostoevsky’s hero in the essay Surrealism: The Last Snapshot of the European Intelligentsia. Stavrogin is characterized by radical ambivalence, embodying the ultimate contradiction between the evil depicted in the crime of violating a child, and the utopic dream of the Golden Age. Benjamin sees Stavrogin as a precursor of surrealism and uses him to justify evil in the revolutionary practice of Surrealists. The first objective is to analyse Benjamin’s essay, to elaborate on how he understands surrealism and why he sees Stavrogin as its predecessor. Further on, I address what are the aesthetic and thematic implications of Benjamin’s reading and why in his concept of Stavrogin images and language precede the self and meaning.     Many approaches have been explored to regenerate the heart muscle followingischemic injury, out of which is the induction of cardiomyocytes proliferation. Ourprevious work demonstrated that cell-cycle was successfully induced incardiomyocytes by viral expression of combination of four cell-cycle factors: cyclinB1, CDK1, cyclin D1 and CDK4; termed as 4 factors (4F). However, only 15-20 % ofthe cells expressing the four factors progress into cell-cycle, while the remainderare quiescent. A pertinent question in the field of cardiac regeneration is why allviral or in vivo transgenic approaches to induce adult cardiomyocyte proliferation,e.g., 4F, YAP, and CyclinA2, promote proliferation in only a subset ofcardiomyocytes. This general finding suggests that factors or conditions beyondcell-cycle induction influence the probability and perpetuation of cardiomyocytedivision.Here we aim to investigate why only a subpopulation of cardiomyocytes is able toprogress through cell-cycle.Temporal single cell RNA sequencing of 60 days mature hiPS-CMs 24, 48 and 72hours post infection with 4F or control virus revealed that a unique population ofcardiomyocytes in the LacZ control group [1026 cells out of 6761 cells (~15%)] thatdisappears after treatment with 4F; another unique cluster with similar cellnumbers appears 24 h after 4F transduction (Cluster 3), which suggests that theinitial population was primed to proliferate. One of the major characteristics of thisprimed subpopulation is the expression of CD36, a major fatty acids transporter incardiomyocytes, and that 4F induction of cell-cycle completely abolishes CD36expression. knocking down CD36 in hiPS-CMs for 48 hours followed by induction ofproliferation using 4F led to 50-70% reduction in proliferation capacity of thecardiomyocytes compared to the control cells. Furthermore, cardiomyocytesisolated at P7 from CD36 knockout mice showed 30-40 % reduction incardiomyocytes proliferation capacity at baseline and after 4F induction ofproliferation compared to cardiomyocytes isolated from WT controls.These findings suggest that CD36 is needed to prime the CMs to proliferate, andthis can be, at least partially, through provision of energy requirements throughβ-oxidation of fatty acids.

    Agriculture, domestic users, and industry rely on water resources systems for fulfilling water demand, while water resources systems face both climate-induced extreme events and management and governance problems. These constraints lead to a mismatch between demand and supply of water for those sectors. This study applies central tendency and variability to analyze data and a mixed methods approach to interpret the result. From 1999 to 2019, the growth rates of population, gross domestic product, and urban population are ­1.354, 6.084, and 3.70%, respectively, contributing to increased water demand. However, the average groundwater depletion increased from 2.455 km3 (1989–1990) to 4.9016 km3, while the average flood-affected areas enhanced by 8,644 km2 in 2014–2018 compared to 1987–1991. Furthermore, salt-affected areas incremented by 222,810 ha in 2009 contrasted to 1973, whereas the mean wind speed of cyclones increased by 30.02 km in 2015–2020 compared to 1988–1995. The mean sea-level rise increased by 16.8 and 169.2 cm in 1995–2000 compared to 1979–1983 in Cox's Bazar and the Char Changa station, respectively. The Mann–Kendall test was applied to detect the trend. This study urges policymakers, water experts, and academics to promote rainwater harvesting that is sustainable to govern rainwater and mitigate water and economic poverty.     Muscular Dystrophies (MDs) are a group of rare inherited genetic muscular pathologies encompassing a variety of clinical phenotypes, gene mutations and mechanisms of disease. MDs undergo progressive skeletal muscle degeneration causing severe health problems that lead to poor life quality, disability and premature death. There are no available therapies to counteract the causes of these diseases and conventional treatments are administered only to mitigate symptoms. Recent understanding on the pathogenetic mechanisms allowed the development of novel therapeutic strategies based on gene therapy, genome editing CRISPR/Cas9 and drug repurposing approaches. Despite the therapeutic potential of these treatments, once the actives are administered, their instability, susceptibility to degradation and toxicity limit their applications. In this frame, the design of delivery strategies based on nanomedicines holds great promise for MD treatments. This review focuses on nanomedicine approaches able to encapsulate therapeutic agents such as small chemical molecules and oligonucleotides to target the most common MDs such as Duchenne Muscular Dystrophy and the Myotonic Dystrophies. The challenge related to in vitro and in vivo testing of nanosystems in appropriate animal models is also addressed. Finally, the most promising nanomedicine-based strategies are highlighted and a critical view in future developments of nanomedicine for neuromuscular diseases is provided.

    AbstractBackgroundBombyx mori silk fibroin is a biomacromolecule that allows the assembly of scaffolds for tissue engineering and regeneration purposes due to its cellular adhesiveness, high biocompatibility and low immunogenicity. Earlier work showed that two types of 3D silk fibroin nonwovens (3D-SFnws) implanted into mouse subcutaneous tissue were promptly vascularized via undefined molecular mechanisms. The present study used nontumorigenic adult human dermal fibroblasts (HDFs) adhering to a third type of 3D-SFnws to assess whether HDFs release exosomes whose contents promote neoangiogenesis.MethodsElectron microscopy imaging and physical tests defined the features of the novel carded/hydroentangled 3D-SFnws. HDFs were cultured on 3D-SFnws and polystyrene plates in an exosome-depleted medium. DNA amounts and D-glucose consumption revealed the growth and metabolic activities of HDFs on 3D-SFnws. CD9-expressing total exosome fractions were from conditioned media of 3D-SFnws and 2D polystyrene plates HDF cultures. Angiogenic growth factors (AGFs) in equal amounts of the two groups of exosomal proteins were analysed via double-antibody arrays. A tube formation assay using human dermal microvascular endothelial cells (HDMVECs) was used to evaluate the exosomes’ angiogenic power.ResultsThe novel features of the 3D-SFnws met the biomechanical requirements typical of human soft tissues. By experimental day 15, 3D-SFnws-adhering HDFs had increased 4.5-fold in numbers and metabolized 5.4-fold more D-glucose than at day 3 in vitro. Compared to polystyrene-stuck HDFs, exosomes from 3D-SFnws-adhering HDFs carried significantly higher amounts of AGFs, such as interleukin (IL)-1α, IL-4 and IL-8; angiopoietin-1 and angiopoietin-2; angiopoietin-1 receptor (or Tie-2); growth-regulated oncogene (GRO)-α, GRO-β and GRO-γ; matrix metalloproteinase-1; tissue inhibitor metalloproteinase-1; and urokinase-type plasminogen activator surface receptor, but lesser amounts of anti-angiogenic tissue inhibitor metalloproteinase-2 and pro-inflammatory monocyte chemoattractant protein-1. At concentrations from 0.62 to 10 μg/ml, the exosomes from 3D-SFnws-cultured HDFs proved their angiogenic power by inducing HDMVECs to form significant amounts of tubes in vitro.ConclusionsThe structural and mechanical properties of carded/hydroentangled 3D-SFnws proved their suitability for tissue engineering and regeneration applications. Consistent with our hypothesis, 3D-SFnws-adhering HDFs released exosomes carrying several AGFs that induced HDMVECs to promptly assemble vascular tubes in vitro. Hence, we posit that once implanted in vivo, the 3D-SFnws/HDFs interactions could promote the vascularization and repair of extended skin wounds due to burns or other noxious agents in human and veterinary clinical settings.

    Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive tumors; it is often diagnosed at an advanced stage and is hardly treatable. These issues are strictly linked to the absence of early diagnostic markers and the low efficacy of treatment approaches. Recently, the study of the metabolic alterations in cancer cells has opened the way to important findings that can be exploited to generate new potential therapies. Within this scenario, mitochondria represent important organelles within which many essential functions are necessary for cell survival, including some key reactions involved in energy metabolism. These organelles remodel their shape by dividing or fusing themselves in response to cellular needs or stimuli. Interestingly, many authors have shown that mitochondrial dynamic equilibrium is altered in many different tumor types. However, up to now, it is not clear whether PDAC cells preferentially take advantage of fusion or fission processes since some studies reported a wide range of different results. This review described the role of both mitochondria arrangement processes, i.e., fusion and fission events, in PDAC, showing that a preference for mitochondria fragmentation could sustain tumor needs. In addition, we also highlight the importance of considering the metabolic arrangement and mitochondria assessment of cancer stem cells, which represent the most aggressive tumor cell type that has been shown to have distinctive metabolic features to that of differentiated tumor cells.
