Here we analysed metagenome-assembled genomes of the “Genomes from Earth’s Microbiomes” initiative from diverse environmental samples, which almost double the known phylogenetic diversity of the phylum. Our data and analyses provide an extended perspective of the variability of chlamydial biology and the ecology of this phylum of intracellular microbes.

Since cocoa trees only grow in some tropical areas, this increased demand have pushed farmers to convert regions of intact forests into cocoa plantations. Between 2001 and 2014 Ghana lost 175,000 hectares of forest due to cocoa expansion. Such amount of deforestation can have a serious impact on biodiversity and climate change. For this reason, food manufacturers have started to develop strategies to replace or reduce the amount of cocoa used in chocolate. One of them is the use of cocoa butter equivalents, mixtures of triglycerides from multiple sources (e.g., sunflower oil, mango kernel, sal) that resemble cocoa butter in physical and chemical properties. In this project we will investigate the crystallization behaviour of selected CBEs from different origins, and compare it with that of cocoa butter. This study can enable the design of more sustainable chocolate recipes and manufacturing processes.

Our initial experiment at ID16B revealed large and unexpected structural anisotropies discovered by analyzing the full 2D diffraction pattern. The patterns showed large intensity variations as a function of the azimuthal angle. The results of the studies we propose on several samples with different growth conditions and degrees of stability can inform the relationship between the observed anisotropies and important properties of metallic glasses, pointing to the need to have structural information beyond the density-density pair distribution function. Further, these studies may provide new information relevant to the glass transition, a long-standing open question in condensed matter physics.

The M plants were grown and sampled in vitro and the St1, St2, St3 plants in the green house. The DMRs were called using the the EpiDiverse/dmr bioinformatic analysis pipeline (Nunn et al., 2021). Stress assays in vitro One-month-old seedlings were transferred to a fresh MS media and growth chambers at 24oC/21oC (day/night),16 h light/8 h dark, as control conditions. For heat-stress, plants were exposed to 30oC (day/night) for one week followed by 2 days of recovery (24oC/21oC) on fresh medium as well as the control plants. Then, the plates were transferred to 37oC (day/night) for 1 week with 2 recovery days (Figure 1A). We sampled aerial parts of plants for the molecular analyses. To reduce variability resulting from individual plants, three biological replicates of 5 pooled plants were collected per condition. Samples were harvested in 1.5 mL tubes between 9:00-11:00 a.m. and immediately frozen in liquid nitrogen and stored at -80oC until required. Greenhouse propagation assays In vitro plants after heat and control treatment were transferred to soil (one plant per pot) in square plastic pots (size: 12x12x10 cm) and to a greenhouse with long day conditions (24oC/21oC day/night and 60%-70% humidity). Twelve mother plants (M) from control (CM; n=12) and heat-stress (HM; n=12) conditions were used for asexual propagation. From each mother plant, the two first stolons (St) were kept for producing the daughter plants of the first asexual propagation (St1) in individual pots. After two weeks, following root formation, the stolons were cut to get independent daughter plants from their mother plant (M). This process was continued until St3. During the campaigns, the integrated CO2 and H2O open-path gas analyzer and 3-D sonic anemometer Irgason by Campbell Scientific (the serial number 1243) was operated on the shore of two glacial lakes. In the first experiment, the Irgason was deployed on the north-west shore of Lake Zub/Priyadarshini at a distance of 10 m from the coast used from 01.01.2018 to 07.02.2018. The instrument was placed on the aluminium tripod on the height of 2 m, and its sensor was directed toward 137° south-eastwards (SE). The second experiment lasted from 07.12.2019 to 13.01.2020; the Irgason was deployed at a distance of 10 m from the shore of Lake Glubokoe at a height of 1.8 m and the sensor was directed toward 144° south-eastwards (SE). The data are stored in the datafiles with the format given in Irgason_output.pdf; the raw data are packed into the *.dat files (one per day) and then compressed (bz2). The calibration of the Irgason was done 21.08.2017 and 13.11.2019 in the lab of the Finnish Meteorological Institute with standard zero-and-span procedure, and then the instrument was adjusted accordingly. The Irgason joined the inter-calibration campaign carried out in October 2018 in Lake Alqueva (Portugal). The work has received financial support from the Academy of Finland (contract number 304345, PI Timo Vihma); the logistic support of the field campaigns provided by the Finnish Antarctic Research Program and the Russian Antarctic Expedition.

Since cocoa trees only grow in some tropical areas, this increased demand have pushed farmers to convert regions of intact forests into cocoa plantations. Between 2001 and 2014 Ghana lost 175,000 hectares of forest due to cocoa expansion. Such amount of deforestation can have a serious impact on biodiversity and climate change. For this reason, food manufacturers have started to develop strategies to replace or reduce the amount of cocoa used in chocolate. One of them is the use of cocoa butter equivalents, mixtures of triglycerides from multiple sources (e.g., sunflower oil, mango kernel, sal) that resemble cocoa butter in physical and chemical properties. In this project we will investigate the crystallization behaviour of selected CBEs from different origins, and compare it with that of cocoa butter. This study can enable the design of more sustainable chocolate recipes and manufacturing processes.

Our initial experiment at ID16B revealed large and unexpected structural anisotropies discovered by analyzing the full 2D diffraction pattern. The patterns showed large intensity variations as a function of the azimuthal angle. The results of the studies we propose on several samples with different growth conditions and degrees of stability can inform the relationship between the observed anisotropies and important properties of metallic glasses, pointing to the need to have structural information beyond the density-density pair distribution function. Further, these studies may provide new information relevant to the glass transition, a long-standing open question in condensed matter physics.
