Abstract Background 3-Hydroxypropionic acid (3-HP) is an important platform chemical, serving as a precursor for a wide range of industrial applications such as the production of acrylic acid and 1,3-propanediol. Although Escherichia coli or Saccharomyces cerevisiae are the primary industrial microbes for the production of 3-HP, alternative engineered hosts have the potential to generate 3-HP from other carbon feedstocks. Methylobacterium extorquens AM1, a facultative methylotrophic α-proteobacterium, is a model system for assessing the possibility of generating 3-HP from one-carbon feedstock methanol. Results Here we constructed a malonyl-CoA pathway by heterologously overexpressing the mcr gene to convert methanol into 3-HP in M. extorquens AM1. The engineered strains demonstrated 3-HP production with initial titer of 6.8 mg/l in shake flask cultivation, which was further improved to 69.8 mg/l by increasing the strength of promoter and mcr gene copy number. In vivo metabolic analysis showed a significant decrease of the acetyl-CoA pool size in the strain with the highest 3-HP titer, suggesting the supply of acetyl-CoA is a potential bottleneck for further improvement. Notably, 3-HP was rapidly degraded after the transition from exponential phase to stationary phase. Metabolomics analysis showed the accumulation of intracellular 3-hydroxypropionyl-CoA at stationary phase with the addition of 3-HP into the cultured medium, indicating 3-HP was first converted to its CoA derivatives. In vitro enzymatic assay and β-alanine pathway dependent 13C-labeling further demonstrated that a reductive route sequentially converted 3-HP-CoA to acrylyl-CoA and propionyl-CoA, with the latter being reassimilated into the ethylmalonyl-CoA pathway. The deletion of the gene META1_4251 encoding a putative acrylyl-CoA reductase led to reduced degradation rate of 3-HP in late stationary phase. Conclusions We demonstrated the feasibility of constructing the malonyl-CoA pathway in M. extorquens AM1 to generate 3-HP. Furthermore, we showed that a reductive route coupled with the ethylmalonyl-CoA pathway was the major channel responsible for degradation of the 3-HP during the growth transition. Engineered M. extorquens AM1 represents a good platform for 3-HP production from methanol.

Following the unsuccessful conclusion in 1930 of M.R. James’s work on the University Library’s medieval manuscripts, the next concerted attempt to produce updated descriptions of the collection was undertaken by Harold Pink, a member of staff in the Library’s Department of Manuscripts. Pink laboured on this task from 1948 until his retirement in 1970, in many instances revising those descriptions produced by James twenty years earlier. He began with those manuscripts in the Additionals classmark sequence, under the guidance of Roger Mynors, who himself contributed a number of descriptions, before later moving on to producing descriptions of manuscripts within the Two-Letter classmark sequence. Both Pink’s and Mynors’ unpublished descriptions have been accessioned into the University Archives as UA ULIB 7/3/75. At some stage, photocopies were made of the entire stock of their handwritten descriptions. These have now been scanned and are made available here for download as pdf files. In most respects, Pink’s and Mynors’ work on the Additional manuscripts has been superseded by that of Jayne Ringrose. Her Summary catalogue of the Additional medieval manuscripts in Cambridge University Library acquired before 1940 was published by Brewer in 2009, but is now also available on Apollo, together with more detailed descriptions of the manuscripts on which the published summary descriptions were based. From the attached plaque: "This kiln is a reminder of the 19th Century when potteries and brickfields were established here amid some of the poorest housing conditions in London; it is one of the few examples of a bottle kiln left in London. In maart 2020 is een archeologisch vooronderzoek uitgevoerd in een plangebied aan de Genderensedijk 7-13 in Genderen (gemeente Altena). De aanleiding van het onderzoek wordt gevormd door het voornemen om voor het plangebied een nieuw bestemmingsplan op te stellen, zodat in de toekomst woningbouw plaats kan vinden. Het archeologisch vooronderzoek bestaat hier uit een gecombineerd onderzoek, te weten een Archeologisch Bureauonderzoek (BO) en een Inventariserend Veldonderzoek (IVO), verkennende fase. Het doel van het archeologisch bureauonderzoek is het specificeren van de archeologische verwachting. Het doel van het inventariserend veldonderzoek is het toetsen en waar mogelijk bijstellen van de gespecificeerde archeologische verwachting, zodat duidelijk wordt of een nieuwe archeologieparagraaf aan het nieuwe bestemmingsplan moet worden toegevoegd.

Within populations of brooding sessile corals, sperm dispersal constitutes the mechanism by which gametes interact and mating occurs, and forms the first link in the network of processes that determine species-wide connectivity patterns. However, almost nothing is known about sperm dispersal for any internally fertilizing coral. In this study, we conducted a parentage analysis on coral larvae collected from an area of mapped colonies, in order to measure the distance sperm disperses for the first time in a reef-building coral and estimated the mating system characteristics of a recently identified putative cryptic species within the Seriatopora hystrix complex (ShA; Warner et al. 2015). We defined consensus criteria among several replicated methods (COLONY 2.0, Cervus 3.0, MLTR v3.2) to maximize accuracy in paternity assignments. Thirteen progeny arrays indicated that this putative species produces exclusively sexually-derived, primarily outcrossed larvae (mean tm=0.999) in multiple paternity broods (mean rp=0.119). Self-fertilization was directly detected at low frequency for all broods combined (2.8%), but comprised 23% of matings in one brood. Although over 82% of mating occurred between colonies within 10 m of each other (mean sperm dispersal = 5.5 m ±4.37 SD), we found no evidence of inbreeding in the established population. Restricted dispersal of sperm compared to slightly greater larval dispersal appears to limit inbreeding among close relatives in this cryptic species. Our findings establish a good basis for further work on sperm dispersal in brooding corals and provide the first information about the mating system of a newly identified and abundant cryptic species.
