    On the basis of two sedimentary records from the central Sea of Okhotsk, we reconstruct the closely coupled glacial/interglacial changes in terrigenous flux, marine productivity, and sea ice coverage over the past 1.1 Myr. The correspondance of our sedimentary records to the China loess grain size record (China loess particle timescale, CHILOPARTS) suggests that environmental changes in both the Sea of Okhotsk area and in SE Asia were closely related via the Siberian atmospheric high-pressure cell. During full glacial times our records point to a strong Siberian High causing northerly wind directions, the extension of the sea ice cover, and a reduced Amur River discharge. Deglacial maxima of terrigenous flux were succeeded by or synchronous to high-productivity events. Marine productivity was strengthened during glacial terminations because of an effective nutrient utilization at times of enhanced water column stratification and high nutrient supply from fluvial runoff and sea ice thawing. During interglacials, SE monsoonal winds prevailed, analogous to today's summer situation of a pronounced Mongolian Heat Low and a strong Hawaiian High. Strong freshwater discharge induced by high precipitation rates in the Amur drainage area and a seasonally reduced and mobile sea ice cover favored marine productivity (although being considerably lower than during the terminations) and a lowered flux of ice-rafted detritus.     Objectives: Natural language processing (NLP) and machine learning approaches were used to build classifiers to identify genomic-related treatment changes in the free-text visit progress notes of cancer patients. Methods: We obtained 5,889 de-identified progress reports (2,439 words on average) for 755 cancer patients who have undergone a clinical Next Generation Sequencing (NGS) testing in Wake Forest Baptist Comprehensive Cancer Center for our data analyses. An NLP system was implemented to process the free-text data and extract NGS-related information. Three types of recurrent neural network (RNN) namely, gated recurrent unit (GRU), long-short term memory (LSTM), and bidirectional LSTM (LSTM_Bi) were applied to classify documents to the treatment-change and no-treatment-change groups. Further, we compared the performances of RNNs to five machine learning algorithms including Naive Bayes (NB), K-nearest Neighbor (KNN), Support Vector Machine for classification (SVC), Random Forest (RF), and Logistic Regression (LR). Results: Our results suggested that, overall, RNNs outperformed traditional machine learning algorithms, and LSTM_Bi showed the best performance among the RNNs in terms of accuracy, precision, recall, and F1 score. In addition, pre-trained word embedding can improve the accuracy of LSTM by 3.4% and reduce the training time by more than 60%. Discussion and Conclusion: NLP and RNN-based text mining solutions have demonstrated advantages in information retrieval and document classification tasks for unstructured clinical progress notes.     The boron isotope composition (d11B) of planktic foraminiferal calcite, which reflects seawater pH, is a well-established proxy for reconstructing palaeo-atmospheric CO2 and seawater carbonate chemistry. However, to translate d11B measurements determined in calcareous fossils into pH we need to know the boron isotope composition of the parent seawater (d11Bsw). While a number of d11Bsw reconstructions exist, the discrepancies between them reveals uncertainties and deficiencies that need to be addressed. Here we present a new d11Bsw record based on the d11B difference between planktic and benthic foraminifera and an estimate of the pH gradient between surface and deep water. We then calculate d11Bsw two different ways. One variant of our method assumes that the pH gradient between surface and deep has remained the same as today over the past 23 Ma; the other uses the d13C gradient between surface and deep to represent change in the pH gradient through time. The results of these two methods of calculating d11Bsw are broadly consistency with each other, however, based on extensive carbon cycle modelling using CYCLOPS and GENIE we favour the d13C gradient method. In our favoured d11Bsw reconstruction, d11Bsw is around 2 per mil lower than today at ~37.5 per mil during the early and middle Miocene and increases to the modern value (39.61 per mil) by ~5 Ma. A similar pattern of change is evident in the seawater composition of three other stable isotope systems, Mg, Li and Ca. Concurrent shifts in the seawater isotopic composition of all four of these elements during the late Miocene, suggest a common forcing mechanism. We hypothesise the most likely cause of these shifts is a change in the isotopic composition of the riverine input, potentially driven by an increase in secondary mineral formation since ~15 Ma. Here we reconcile the conflicting evidence by proposing that the spread of agriculture involved a life history quality–quantity trade-off whereby mothers traded offspring survival for increased fertility, achieving greater reproductive success despite deteriorating health. We test this hypothesis by investigating fertility, mortality, health, and overall reproductive success in Agta hunter-gatherers whose camps exhibit variable levels of sedentarization, mobility, and involvement in agricultural activities. We conducted blood composition tests in 345 Agta and found that viral and helminthic infections as well as child mortality rates were significantly increased with sedentarization. Nonetheless, both age-controlled fertility and overall reproductive success were positively affected by sedentarization and participation in cultivation. Thus, we provide the first empirical evidence, to our knowledge, of an adaptive mechanism in foragers that reconciles the decline in health and child survival with the observed demographic expansion during the Neolithic.

    We freeze-dried and homogenized 44 samples of c. 0.7-1.8 g dry sediment from core PG1351 covering late glacials and interglacials of MIS 8 to MIS 5e, integrating sediment of 1 cm core depth. Temporal resolution of these samples ranges from 140 to 960 years per sample. For the period between 430 and 405 kyrs ago (end of MIS 12 to MIS 11c), 13 samples of 0.5-1.3 g of dry sediment from ICDP core 5011-1 were taken for MA analyses, integrating sediment of 2 cm core depth. Eight of these 13 samples are from the same core depths as were previously analysed for pollen (Melles et al., 2012). Temporal resolution of these samples varies between 200 and 970 years per sample comparable to core PG1351. Across all samples, temporal resolution is 333 ± 273 years per sample, giving centennial- to millennial scale averages.We extracted the polar lipids of all MA samples using a Dionex Accelerated Solvent Extraction system (ASE 350, ThermoFisher Scientific) at 100°C, 103 bar pressure and two extraction cycles (20 min static time) with 100 % methanol, after an ASE cycle with 100 % dichloromethane. For every sample sequence (n=13-18), we extracted a blank ASE cell and included it in all further steps. We added 60 ng of deuterated levoglucosan (C6H3D7O5; dLVG; Th. Geyer GmbH & Co. KG) as internal standard, and filtered the extract over a PTFE filter using acetonitrile and 5 % HPLC-grade water. We analysed the extracts with an Ultimate 3000 RS ultra-high performance liquid chromatograph (U-HPLC) with thermostated autosampler and column oven coupled to a Q Exactive Plus Orbitrap mass spectrometer (Quadrupole-Orbitrap MS; ThermoFisher Scientific) with heated electrospray injection (HESI) probe at GFZ Potsdam, using measurement conditions adapted from earlier studies (Hopmans et al., 2013;Schreuder et al., 2018;Dietze et al., 2019). Briefly, separation was achieved on two Xbridge BEH amide columns in series (2.1 x 150 mm, 3.5 um particle size) fitted with a 50 mm pre-column of the same material (Waters). The compounds were eluted (flow rate 0.2 mL min-1) with 100 % A for 15 minutes, followed by column cleaning with 100 % B for 15 min, and re-equilibration to starting conditions for 25 min. Eluent A was acetonitrile:water:triethylamine (92.5:7.5:0.01) and eluent B acetonitrile:water:triethylamine (70:30:0.01). HESI settings were as follows: sheath gas (N2) pressure 20 (arbitrary units), auxiliary gas (N2) pressure 3 (arbitrary units), auxiliary gas (N2) temperature of 50 ˚C, spray voltage -2.9 kV (negative ion mode), capillary temperature 300 °C, S-Lens 50 V. Detection was achieved by monitoring m/z 150-200 with a resolution of 280,000 ppm. Targeted data dependent MS2 (normalized collision energy 13 V) was performed on any signal within 10 ppm of m/z 161.0445 (calculated exact mass of deprotonated levoglucosan and its isomers) or m/z 168.0884 (calculated exact mass of deprotonated dLVG) with an isolation window of 0.4 m/z. The detection limit was 2.5 pg on column, based on injections of 0.5 to 5000 pg on column of authentic standards of LVG, MAN, and GAL (Santa Cruz Biotechnology) and dLVG.Integrations were performed on mass chromatograms within 3 ppm mass accuracy and corrected for relative response factors to dLVG (1.08 ± 0.10, 0.76 ± 0.10 and 0.24 ± 0.05 for LVG, MAN, and GAL, respectively), according to known authentic standard mixes injected before and after every measurement sequence and supported by characteristic isomer-specific MS² data. All samples were corrected by subtracting the maximum MA concentrations in the blank duplicates of each ASE sequence. To account for biases due to sediment properties and sedimentation rates, MA influxes (mass accumulation rates in ng cm-2 yr-1) were calculated by multiplying the concentrations (ng g-1) with the sample-specific dry bulk densities (Melles et al., 2007;Wennrich et al., 2016), and the sample's sedimentation rates (cm yr-1) using the age-depth models presented by Nowaczyk et al. (2013) for the the PG1351 and the ICDP-5011-1 cores.

    The combination of permafrost history and dynamics, lake level changes and the tectonical framework is considered to play a crucial role for sediment delivery to El'gygytgyn Crater Lake, NE Russian Arctic. The purpose of this study is to propose a depositional framework based on analyses of the core strata from the lake margin and historical reconstructions from various studies at the site. A sedimentological program has been conducted using frozen core samples from the 141.5 m long El'gygytgyn 5011-3 permafrost well. The drill site is located in sedimentary permafrost west of the lake that partly fills the El'gygytgyn Crater. The total core sequence is interpreted as strata building up a progradational alluvial fan delta. Four macroscopically distinct sedimentary units are identified. Unit 1 (141.5-117.0 m) is comprised of ice-cemented, matrix-supported sandy gravel and intercalated sandy layers. Sandy layers represent sediments which rained out as particles in the deeper part of the water column under highly energetic conditions. Unit 2 (117.0-24.25 m) is dominated by ice-cemented, matrix-supported sandy gravel with individual gravel layers. Most of the Unit 2 diamicton is understood to result from alluvial wash and subsequent gravitational sliding of coarse-grained (sandy gravel) material on the basin slope. Unit 3 (24.25-8.5 m) has ice-cemented, matrix-supported sandy gravel that is interrupted by sand beds. These sandy beds are associated with flooding events and represent near-shore sandy shoals. Unit 4 (8.5-0.0 m) is ice-cemented, matrix-supported sandy gravel with varying ice content, mostly higher than below. It consists of slope material and creek fill deposits. The uppermost metre is the active layer (i.e. the top layer of soil with seasonal freeze and thaw) into which modern soil organic matter has been incorporated. The nature of the progradational sediment transport taking place from the western and northern crater margins may be related to the complementary occurrence of frequent turbiditic layers in the central lake basin, as is known from the lake sediment record. Slope processes such as gravitational sliding and sheet flooding occur especially during spring melt and promote mass wasting into the basin. Tectonics are inferred to have initiated the fan accumulation in the first place and possibly the off-centre displacement of the crater lake.     On the basis of two sedimentary records from the central Sea of Okhotsk, we reconstruct the closely coupled glacial/interglacial changes in terrigenous flux, marine productivity, and sea ice coverage over the past 1.1 Myr. The correspondance of our sedimentary records to the China loess grain size record (China loess particle timescale, CHILOPARTS) suggests that environmental changes in both the Sea of Okhotsk area and in SE Asia were closely related via the Siberian atmospheric high-pressure cell. During full glacial times our records point to a strong Siberian High causing northerly wind directions, the extension of the sea ice cover, and a reduced Amur River discharge. Deglacial maxima of terrigenous flux were succeeded by or synchronous to high-productivity events. Marine productivity was strengthened during glacial terminations because of an effective nutrient utilization at times of enhanced water column stratification and high nutrient supply from fluvial runoff and sea ice thawing. During interglacials, SE monsoonal winds prevailed, analogous to today's summer situation of a pronounced Mongolian Heat Low and a strong Hawaiian High. Strong freshwater discharge induced by high precipitation rates in the Amur drainage area and a seasonally reduced and mobile sea ice cover favored marine productivity (although being considerably lower than during the terminations) and a lowered flux of ice-rafted detritus.
