    Abstract Background Intermittent hemodialysis (IHD) is the modality of choice in the extracorporeal treatment (ECTR) of acute methanol poisoning. However, the comparative clinical effectiveness of intermittent versus continuous modalities (CRRT) is unknown. During an outbreak of mass methanol poisoning, we therefore studied the effect of IHD versus CRRT on mortality and the prevalence of visual/central nervous system (CNS) sequelae in survivors. Methods The study was designed as prospective observational cohort study. Patients hospitalized with a diagnosis of acute methanol poisoning were identified for the study. Exploratory factor analysis and multivariate logistic regression were applied to determine the effect of ECTR modality on the outcome. Results Data were obtained from 41 patients treated with IHD and 40 patients with CRRT. The follow-up time in survivors was two years. Both groups of patients were comparable by age, time to presentation, laboratory data, clinical features, and other treatment applied. The CRRT group was more acidemic (arterial blood pH 6.96 ± 0.08 vs. 7.17 ± 0.07; p  0.05). Conclusions In spite of the faster correction of the acidosis and the quicker removal of the toxic metabolite in intermittent dialysis, we did not find significant differences in the treatment outcomes between the two groups after adjusting for the degree of acidemia and the severity of poisoning on admission. These findings support the strategy of “use what you have” in situations with large outbreaks and limited dialysis capacity.

    Cytokinins represent an important group of plant growth regulators that can modulate several biotechnological processes owing to their ability to influence almost all stages of plant development and growth. In addition, the use of purine based cytokinins with aromatic substituent in C6 position of the purine moiety in tissue culture techniques is currently experiencing a surge in interest, made possible by the ongoing systematic synthesis and study of these compounds. This review article outlines progress in the synthesis of aromatic cytokinins, the in vitro and in vivo effects of these substances and insights gleaned from their synthesis. As the purine moiety in these compounds can be substituted at several positions, we examine each of the substitution possibilities in relation to the derivatives prepared so far. The discussion highlights the gradual simplification of their preparation in relation to their application in practice and summarizes the relevant organic chemistry literature and published patents.

    A relationship between the heliospheric magnetic field, atmospheric electric field, lightning activity, and secondary cosmic rays measured on the high mount of Lomnický Štít (2,634 m a.s.l.), Slovakia, during the declining phase of the solar cycle 24 is investigated with a focus on variations related to solar rotation (about 27 days). The secondary cosmic rays are detected using a neutron monitor and the detector system SEVAN, which distinguishes between different particles and energies. Using spectral analysis, we found distinct ∼27-day periodicities in variations of Bx and By components of the heliospheric magnetic field and in pressure-corrected measurements of secondary cosmic rays. The 27-day variations of secondary cosmic rays, on average, advanced and lagged the variations of Bx and By components by about 40° and −140°, respectively. Distinct 27-day periodicities were found both in the neutron monitor and the SEVAN upper and middle detector measurements. A nondominant periodicity of ∼27 days was also found for lightning activity. A cross-spectral analysis between fluctuation of the lightning activity and fluctuation of the heliospheric magnetic field (HMF) showed that fluctuation of the lightning activity was in phase and in antiphase with Bx and By components of the HMF, respectively, which is in agreement with previous studies investigating the influence of solar activity on lightning. On the other hand, the ∼27-day periodicity was not significant in the atmospheric electric field measured in Slovakia and Czechia. Therefore, no substantial influence of Bx and By on the atmospheric electric field was observed at these middle-latitude stations.

    The paper deals with experimental research of the initial phase of free-settling spherical particle which density is close to the density of water. The experiments were carried out with a smooth particle as well as with a particle with a rough surface. The obtained results show that in the initial phase of sedimentation, there is a formation of vortex structures, which are reflected in the increase of resistance in coefficient values.

    We consider a theory of centers and homotopy centers of monoids in monoidal categories which themselves are enriched in duoidal categories. Duoidal categories (introduced by Aguillar and Mahajan under the name 2-monoidal categories) are categories with two monoidal structures which are related by some, not necessary invertible, coherence morphisms. Centers of monoids in this sense include many examples which are not `classical.' In particular, the 2-category of categories is an example of a center in our sense. Examples of homotopy center (analogue of the classical Hochschild complex) include the Gray-category Gray of 2-categories, 2-functors and pseudonatural transformations and Tamarkin's homotopy 2-category of dg-categories, dg-functors and coherent dg-transformations. 52 pages

    A nucleoside bearing a solvatochromic push–pull fluorene fluorophore (dCFL) was designed and synthesized by the Sonogashira coupling of alkyne-linked fluorene 8 with 5-iodo-2′-deoxycytidine. The fluorene building block 8 and labeled nucleoside dCFL exerted bright fluorescence with significant solvatochromic effect providing emission maxima ranging from 421 to 544 nm and high quantum yields even in highly polar solvents, including water. The solvatochromism of 8 was studied by DFT and ADC(2) calculations to show that, depending on the polarity of the solvent, emission either from the planar or the twisted conformation of the excited state can occur. The nucleoside was converted to its triphosphate variant dCFLTP which was found to be a good substrate for DNA polymerases suitable for the enzymatic synthesis of oligonucleotide or DNA probes by primer extension or PCR. The fluorene-linked DNA can be used as fluorescent probes for DNA–protein (p53) or DNA–lipid interactions, exerting significant color changes visible even to the naked eye. They also appear to be suitable for time-dependent fluorescence shift studies on DNA, yielding information on DNA hydration and dynamics. Color-changing fluorescent nucleotide and oligonucleotide probes for studying interactions with other biomolecules were designed and prepared, and perform better than currently known environment-sensitive fluorophores.

    Abstract Background and Aims Polyploidy is an important contributor to sympatric speciation and assortative mating is a key mechanism driving cytotype interactions in contact zones. While strong reproductive barriers can mediate the coexistence of different cytotypes in sympatry, positive frequency-dependent mating disadvantage ultimately drives the transition to single-ploidy populations. However, comprehensive estimates of reproductive isolation among cytotypes and across multiple barriers are rare. We quantify the strength of isolation across multiple reproductive stages in a tetraploid–octoploid contact zone to understand the potential for coexistence. Methods Assortative mating due to flowering asynchrony, pollinator behaviour, morphological overlap, self-fertilization and gametic competition between tetraploid and octoploid Gladiolus communis in a contact zone in the Western Iberian Peninsula were assessed in natural and experimental populations to quantify reproductive isolation (RI) between cytotypes. Key Results Tetraploids and octoploids have a high degree of overlap in flowering time and similar floral morphology, and are visited by generalist insects without cytotype foraging preferences, resulting in weak pre-pollination RI (from 0.00 to 0.21). In contrast, post-pollination isolation resulting from gametic selection was a strong barrier to inter-cytotype mating, with ploidy composition in stigmatic pollen loads determining the levels of RI (from 0.54 to 1.00). Between-cytotype cross-incompatibility was relatively high (RI from 0.54 to 0.63) as was isolation acquired through self-pollination (RI of 0.59 in tetraploids and 0.39 in octoploids). Conclusions Total RI was high for both tetraploids (from 0.90 to 1.00) and octoploids (from 0.78 to 0.98). Such high rates of assortative mating will enable cytotype coexistence in mixed-ploidy populations by weakening the impacts of minority cytotype exclusion. This study reveals the key role of gametic selection in cytotype siring success and highlights the importance of comprehensive estimates across multiple reproductive barriers to understand cytotype interactions at contact zones.

    Instrumentation for flow cytometry and sorting is designed around the assumption that samples are single-cell suspensions. However, with few exceptions, higher plants comprise complex multicellular tissues and organs, in which the individual cells are held together by shared cell walls. Single-cell suspensions can be obtained through digestion of the cells walls and release of the so-called protoplasts (plants without their cell wall). Here we describe best practices for protoplast preparation, and for analysis through flow cytometry and cell sorting. Finally, the numerous downstream applications involving sorted protoplasts are discussed.

    Abstract The Double Asteroid Redirection Test (DART) mission will be the first test of a kinetic impactor as a means of planetary defense. In late 2022, DART will collide with Dimorphos, the secondary in the Didymos binary asteroid system. The impact will cause a momentum transfer from the spacecraft to the binary asteroid, changing the orbit period of Dimorphos and forcing it to librate in its orbit. Owing to the coupled dynamics in binary asteroid systems, the orbit and libration state of Dimorphos are intertwined. Thus, as the secondary librates, it also experiences fluctuations in its orbit period. These variations in the orbit period are dependent on the magnitude of the impact perturbation, as well as the system’s state at impact and the moments of inertia of the secondary. In general, any binary asteroid system whose secondary is librating will have a nonconstant orbit period on account of the secondary’s fluctuating spin rate. The orbit period variations are typically driven by two modes: a long period and a short period, each with significant amplitudes on the order of tens of seconds to several minutes. The fluctuating orbit period offers both a challenge and an opportunity in the context of the DART mission. Orbit period oscillations will make determining the post-impact orbit period more difficult but can also provide information about the system’s libration state and the DART impact.

    Experimental analyses of depth distributions of phase-specific residual stresses after deep rolling were carried out by means of laboratory X-ray diffraction and neutron diffraction for the two duplex steels X2CrNiMoN22-5-3 and X3CrNiMoN27-5-2, which differ significantly in their ferrite to austenite ratios. The aim of the investigation was to elucidate to which extent comparable results can be achieved with the destructive and the non-destructive approach and how the process induced phase-specific micro residual stresses influence the determination of the phase- and {hkl}-specific reference value d0, required for evaluation of neutron strain scanning experiments. A further focus of the work was the applicability of correction approaches that were developed originally for single-phase materials for accounting for spurious strains during through surface neutron scanning experiments on coarse two-phase materials. The depth distributions of macro residual stresses were separated from the phase-specific micro residual stresses. In this regard, complementary residual stress analysis was carried out by means of incremental hole drilling. The results indicate that meaningful macro residual stress depth distributions can be determined non-destructively by means of neutron diffraction for depths starting at about 150–200 µm. Furthermore, it was shown that the correction of the instrumental surface effects, which are intrinsic for surface neutron strain scanning, through neutron ray-tracing simulation is applicable to multiphase materials and yields reliable results. However, phase-specific micro residual stresses determined by means of neutron diffraction show significant deviations to data determined by means of lab X-ray stress analysis according to the well-known sin2ψ-method.

    Abstract Background Intermittent hemodialysis (IHD) is the modality of choice in the extracorporeal treatment (ECTR) of acute methanol poisoning. However, the comparative clinical effectiveness of intermittent versus continuous modalities (CRRT) is unknown. During an outbreak of mass methanol poisoning, we therefore studied the effect of IHD versus CRRT on mortality and the prevalence of visual/central nervous system (CNS) sequelae in survivors. Methods The study was designed as prospective observational cohort study. Patients hospitalized with a diagnosis of acute methanol poisoning were identified for the study. Exploratory factor analysis and multivariate logistic regression were applied to determine the effect of ECTR modality on the outcome. Results Data were obtained from 41 patients treated with IHD and 40 patients with CRRT. The follow-up time in survivors was two years. Both groups of patients were comparable by age, time to presentation, laboratory data, clinical features, and other treatment applied. The CRRT group was more acidemic (arterial blood pH 6.96 ± 0.08 vs. 7.17 ± 0.07; p  0.05). Conclusions In spite of the faster correction of the acidosis and the quicker removal of the toxic metabolite in intermittent dialysis, we did not find significant differences in the treatment outcomes between the two groups after adjusting for the degree of acidemia and the severity of poisoning on admission. These findings support the strategy of “use what you have” in situations with large outbreaks and limited dialysis capacity.
