Through its actions and with the motto « Together we win, divided we are slow! » several joint research projects in the fields of perioperative medicine, fluid therapy, cardiovascular monitoring and support have been conducted. In the light of the COVID- 19 pandemic, the SepsEast community is aware of its mission and is ready to take the challenge. This is mirrored by several educational, clinical and research activities including the development of a COVID-19 Registry ; and an observational clinical study on cytokine adsorption in COVID-19 patients. The current pandemic should be our lesson on how to manage the global threat of infectious disease and to develop strategies for effective diagnostic and therapeutic procedures. Hopefully, the SepsEast community will contribute to these developments and scientific advances in general.

Galapagos- AbbVie identified a novel potentiator GLPG1837 by compound screening on mutant CFTR. YFP-halide efflux assays and single channel measurements showed ∼2.5-fold improvement in channel activity by GLPG1837 compared to VX-770 (ivacaftor/Kalydeco) on G551D CFTR (1, 2). GLPG1837 successfully passed the Phase-2 clinical trials and proved to be the first potentiator after VX-770 to show competitive results on G551D patients. To identify potential differences in the mode of actions of these potentiators we studied their effects on CFTR folding and function. Biochemical radiolabeling experiments showed that mutations in the intracellular loop 2 (ICL2) disrupt domain assembly between TMD1 and NBD2, a late folding event in CFTR, but in most cases do not impair CFTR trafficking towards the cell surface. Protease-susceptibility assays showed that VX-770 improved late TMD1 folding of many ICL2 mutations, but GLPG1837 did not. YFP-halide efflux assays showed that these ICL2 mutants had varying effect on channel function, ranging from wild-type-like to function-defective mutants. GLPG1837 restored function of non-CF gating mutant E267K much better than VX-770. Residue E267 in ICL2 electrostatically interacts with K1060 in ICL4 to promote channel opening (3). This indicates that GLPG1837 is more efficient in compensating for this lost interaction. Altogether, our biochemical and functional data suggests that potentiators VX-770 and GLPG1837 have a different mode of action.

Bahamonde-Birke; Stefan Trommer; Barbara Lenz;Autonomous driving is being discussed as a promising solution for transportation-related issues and might bring some improvement for users of the system. For instance, especially high mileage commuters might compensate for some of their time spent traveling as they will be able to undertake other activities while going to work. At the same time, there are still many uncertainties and little empirical data on the impact of autonomous driving on mode choices. This study addresses the impact of autonomous driving on value of travel time savings (VTTS) and mode choices for commuting trips using stated-choice experiments. Two use cases were addressed – a privately owned, and a shared autonomous vehicle – compared with other modes of transportation. The collected data were analyzed by performing a mixed logit model. The results show that mode-related factors such as time elements, especially in-vehicle time and cost, play a crucial role for mode choices that include autonomous vehicles. The study provides empirical evidence that autonomous driving may lead to a reduction in VTTS for commuting trips. It was found that driving autonomously in a privately owned vehicle might reduce the VTTS by 31% compared with driving manually, and is perceived similarly to in-vehicle time in public transportation. Furthermore, riding in a shared autonomous vehicle is perceived 10% less negatively than driving manually. The study provides important insights into VTTS by autonomous driving for commuting trips and could be a base for future research to build upon.

As we become more integrated into a global world, technological advances and teaching innovation that are grounded in Science have become crucial. Rapid advancements in science education and information technology provide promising resources that require many academic disciplines to work together. Developing new tools and defining new methodologies to share educational experiences, including empirical studies that support their efficiency, constitute a promising approach to improve Health Sciences. The aim of this session is to encourage and enable the exchange of information related with the advance and support of Health Science Education. In this paper the authors summarize the recent advances in technological innovations in biomedical training and practice. Most of the main trends in this field are reviewed, including: training in health sciences through a variety of resources such as computer simulations, stereoscopic visualization systems with augmented reality glasses, computer platforms for managing and using resources and documents; the generation of three-dimensional images developed with commercial software for 3D reconstruction; medical and surgical simulation using Virtual Reality (RV) and Augmented Reality (AR); the role of stereoscopic vision systems in the health sciences; and the use of teaching medical material reconstructed with 3D printers.

Genome-wide association studies (GWAS) have identified hundreds of loci associated with Crohns disease (CD), however, as with all complex diseases, deriving pathogenic mechanisms from these non-coding GWAS discoveries has been challenging. To complement GWAS and better define actionable biological targets, we analysed sequenced data from more than 30,000 CD patients and 80,000 population controls. We observe rare coding variants in established CD susceptibility genes as well as ten genes where coding variation directly implicates the gene in disease risk for the first time.

Through its actions and with the motto « Together we win, divided we are slow! » several joint research projects in the fields of perioperative medicine, fluid therapy, cardiovascular monitoring and support have been conducted. In the light of the COVID- 19 pandemic, the SepsEast community is aware of its mission and is ready to take the challenge. This is mirrored by several educational, clinical and research activities including the development of a COVID-19 Registry ; and an observational clinical study on cytokine adsorption in COVID-19 patients. The current pandemic should be our lesson on how to manage the global threat of infectious disease and to develop strategies for effective diagnostic and therapeutic procedures. Hopefully, the SepsEast community will contribute to these developments and scientific advances in general.

Galapagos- AbbVie identified a novel potentiator GLPG1837 by compound screening on mutant CFTR. YFP-halide efflux assays and single channel measurements showed ∼2.5-fold improvement in channel activity by GLPG1837 compared to VX-770 (ivacaftor/Kalydeco) on G551D CFTR (1, 2). GLPG1837 successfully passed the Phase-2 clinical trials and proved to be the first potentiator after VX-770 to show competitive results on G551D patients. To identify potential differences in the mode of actions of these potentiators we studied their effects on CFTR folding and function. Biochemical radiolabeling experiments showed that mutations in the intracellular loop 2 (ICL2) disrupt domain assembly between TMD1 and NBD2, a late folding event in CFTR, but in most cases do not impair CFTR trafficking towards the cell surface. Protease-susceptibility assays showed that VX-770 improved late TMD1 folding of many ICL2 mutations, but GLPG1837 did not. YFP-halide efflux assays showed that these ICL2 mutants had varying effect on channel function, ranging from wild-type-like to function-defective mutants. GLPG1837 restored function of non-CF gating mutant E267K much better than VX-770. Residue E267 in ICL2 electrostatically interacts with K1060 in ICL4 to promote channel opening (3). This indicates that GLPG1837 is more efficient in compensating for this lost interaction. Altogether, our biochemical and functional data suggests that potentiators VX-770 and GLPG1837 have a different mode of action.

Bahamonde-Birke; Stefan Trommer; Barbara Lenz;Autonomous driving is being discussed as a promising solution for transportation-related issues and might bring some improvement for users of the system. For instance, especially high mileage commuters might compensate for some of their time spent traveling as they will be able to undertake other activities while going to work. At the same time, there are still many uncertainties and little empirical data on the impact of autonomous driving on mode choices. This study addresses the impact of autonomous driving on value of travel time savings (VTTS) and mode choices for commuting trips using stated-choice experiments. Two use cases were addressed – a privately owned, and a shared autonomous vehicle – compared with other modes of transportation. The collected data were analyzed by performing a mixed logit model. The results show that mode-related factors such as time elements, especially in-vehicle time and cost, play a crucial role for mode choices that include autonomous vehicles. The study provides empirical evidence that autonomous driving may lead to a reduction in VTTS for commuting trips. It was found that driving autonomously in a privately owned vehicle might reduce the VTTS by 31% compared with driving manually, and is perceived similarly to in-vehicle time in public transportation. Furthermore, riding in a shared autonomous vehicle is perceived 10% less negatively than driving manually. The study provides important insights into VTTS by autonomous driving for commuting trips and could be a base for future research to build upon.
