    Abstract. Oxygen optode measurements on floats and gliders suffer from a slow time response and sensors that are or eventually run out of calibration. Based on two dual-O2 Argo floats, we show how to post-correct for the effect of the optode’s time response and give an update on optode in-situ drift stability and in-air calibration. Both floats are equipped with an unpumped Aanderaa 4330 optode and a pumped Sea-Bird SBE63 optode. Response times for the pumped SBE63 were derived after Bittig et al. (2014). Also their methods were used to correct the time response bias. Using both optodes on each float, the time response regime of the unpumped Aanderaa optode was characterized more accurately than previously possible. Response times for the pumped SBE63 are in the range of 25–40 s, while they are between 60–95 s for the unpumped 4330 optode. Our parameterization can be employed to post-correct the slow optode time response on floats and gliders. After correction, both sensors agree to within 2–3 μmol kg−1 (median difference) in the strongest gradients (120 μmol kg−1 change over 8 minutes or 20 dbar) and better elsewhere. However, time response correction is only possible if measurement times are known, i.e., provided by the platform as well as transmitted and stored with the data. The O2 in-air measurements show a significant in-situ optode drift of −0.4 % yr−1 and −0.2 % yr−1, respectively. Optode in-air measurements are systematically biased high during mid-day surfacings compared to dusk, dawn, and nighttime. While preference can be given to nighttime surfacings to avoid this in-air calibration bias, we suggest a parameterization of the daytime effect as function of the sun’s elevation to be able to use all data and to better constrain the result. Taking all effects into account, calibration factors have an uncertainty of 0.1 %. In addition, in-air calibration factors vary by 0.1–0.2 % when using different reanalysis models as reference. The overall accuracy that can be achieved following the proposed correction routines is <1 μmol kg−1.

    ABSTRACT The cell shape of Trypanosoma brucei is influenced by flagellum-to-cell-body attachment through a specialised structure – the flagellum attachment zone (FAZ). T. brucei exhibits numerous morphological forms during its life cycle and, at each stage, the FAZ length varies. We have analysed FLAM3, a large protein that localises to the FAZ region within the old and new flagellum. Ablation of FLAM3 expression causes a reduction in FAZ length; however, this has remarkably different consequences in the tsetse procyclic form versus the mammalian bloodstream form. In procyclic form cells FLAM3 RNAi results in the transition to an epimastigote-like shape, whereas in bloodstream form cells a severe cytokinesis defect associated with flagellum detachment is observed. Moreover, we demonstrate that the amount of FLAM3 and its localisation is dependent on ClpGM6 expression and vice versa. This evidence demonstrates that FAZ is a key regulator of trypanosome shape, with experimental perturbations being life cycle form dependent. An evolutionary cell biology explanation suggests that these differences are a reflection of the division process, the cytoskeleton and intrinsic structural plasticity of particular life cycle forms. Summary: Trypanosoma brucei FLAM3 is a flagellar FAZ protein. Its depletion leads to a reduction in FAZ length, which has different consequences depending on the life cycle stage of the parasite.

    International audience; SIRT1 regulates energy homeostasis by controlling the acetylation status and activity of a number of enzymes and transcriptional regulators. The fact that NAD(+) levels control SIRT1 activity confers a hypothetical basis for the design of new strategies to activate SIRT1 by increasing NAD(+) availability. Here we show that the deletion of the poly(ADP-ribose) polymerase-1 (PARP-1) gene, encoding a major NAD(+)-consuming enzyme, increases NAD(+) content and SIRT1 activity in brown adipose tissue and muscle. PARP-1(-)(/-) mice phenocopied many aspects of SIRT1 activation, such as a higher mitochondrial content, increased energy expenditure, and protection against metabolic disease. Also, the pharmacologic inhibition of PARP in vitro and in vivo increased NAD(+) content and SIRT1 activity and enhanced oxidative metabolism. These data show how PARP-1 inhibition has strong metabolic implications through the modulation of SIRT1 activity, a property that could be useful in the management not only of metabolic diseases, but also of cancer.

    Hydrogel thin films containing temperature sensitive chemical functionalities (such as N-isopropylacrylamide, NIPAAm) are particularly interesting for sensor and actuator setups. Complex 3D structures can be conformally coated by the solvent free technique initiated Chemical Vapor Deposition, with precise control over chemical composition and film thickness. In this study, NIPAAm-based thin films with film thicknesses ranging from tens to several hundreds of nanometers and with different amounts of cross-linking were deposited. Above the lower critical solution temperature (LCST), these films repel out water and hence shrink. The amount of cross-linking and the deposited film thickness were successfully identified to both affect shape and position of the LCST transition of these systems: a promising basis for tuning response properties.

    AbstractComplexes of palladium bearing NHC ligands (NHC=N‐heterocyclic carbene) were synthesized and fully characterized. The [Pd(NHC)(allyl)Cl] series was obtained by simple cleavage of [Pd(allyl)Cl]2 by using either the isolated free NHC or the in situ generated NHC. The NHC ligand sterics were varied by introduction of groups attached to the C4–C5 carbon atoms of the NHC backbone. Results of the X‐ray diffraction study permitted determination of the NHC steric parameters within this series. It was observed that the NHC backbone induces variations in its percent buried volume (%Vbur) as a function of the N‐substituents.(© Wiley‐VCH Verlag GmbH & Co. KGaA, 69451 Weinheim, Germany, 2009)

    This contribution reports on the optimization of the multicomponent CEM II/C-M (S-LL) cement, a cement type which is intended to be introduced with a revised version of the EN 197-1. The replacement of Portland cement clinker has been maximized by a combination of granulated blast-furnace slag and limestone while meeting an adequate cement performance and minimizing its environmental impact.

    Scientific interest in iron-oxides and in particular magnetite has been renewed due to the broad scope of their fascinating properties, which are finding applications in electronics and biomedicine. Specifically, iron oxide nanoparticles (IONPs) are gathering attraction in biomedicine. Their cores are usually constituted by a mixture of maghemite and magnetite phases. In view of this, to fine-tune the properties of an ensemble of IONPs towards their applications, it is essential to enhance mass fabrication processes towards the production of monodisperse IONPs with controlled size, shape, and stoichiometry. We exploit the vacancy sensitivity of the Verwey transition to detect the presence of magnetite. Here we provide direct evidence for the Verwey transition in an ensemble of IONPs through neutron diffraction. This transition is observed as a variation in the Fe magnetic moment at octahedral sites and, in turn, gives rise to a change of the net magnetic moment. Finally, we show this variation as the microscopic ingredient driving the characteristic kink that hallmarks the Verwey transition in thermal variation of magnetization.     AbstractIndividuals and communities socially construct risk, and societies with greater risk perception may be more apt to mobilize or adapt to emergent threats like climate change. Increasing climate change awareness is often considered necessary in the first stages of the adaptation process to manage its impacts and reduce overall vulnerability. Since agriculture is affected by climate change in several ways, farmers can provide first-hand observations of climate change impacts and adaptation options. This paper aims to identify the current research trends and set the future research agenda on climate change awareness, perceived impacts, and adaptive capacity from farmers’ experiences and behavior. We analyzed a portfolio of 435 articles collected from WoS and Scopus databases between 2010 and 2020 using bibliometrics. From the original portfolio, we select 108 articles for a more comprehensive and systematic review. Publication trends and content analysis have been employed to identify influential work, delineate the mental structure of farmers’ beliefs and concerns, and identify main research gaps. The comprehensive analysis reported (1) farmers’ socio-demographic characteristics influencing farmers’ perceptions; (2) awareness and changing climate evidence due to human activity; (3) the main perceived effects (rising temperatures, changing rainfall patterns, and extreme events); (4) the most relevant adaptation measures (crop changing and soil/water conservation techniques); and (5) factors and barriers limiting adaptation (lack of information, credit, and expertness). The review outlines the main gaps and their drivers to help future researchers, managers, and decision-makers to prioritize their actions according to farmers’ concerns and their adaptive capacity to reduce farming vulnerability.

    The family Gemmataceae accommodates aerobic, chemoorganotrophic planctomycetes, which inhabit various freshwater ecosystems, wetlands and soils. Here, we describe a novel member of this family, strain PX52T, which was isolated from a boreal eutrophic lake in Northern Russia. This isolate formed pink-pigmented colonies and was represented by spherical cells that occurred singly, in pairs or aggregates and multiplied by budding. Daughter cells were highly motile. PX52T was an obligate aerobic chemoorganotroph, which utilized various sugars and some heteropolysaccharides. Growth occurred at pH 5.0–7.5 (optimum pH 6.5) and at temperatures between 10 and 30 °C (optimum 20–25 °C). The major fatty acids were C18 : 1ɷ7c, C18 : 0 and βOH-C16:0; the major intact polar lipid was trimethylornithine, and the quinone was MK-6. The complete genome of PX52T was 9.38 Mb in size and contained nearly 8000 potential protein-coding genes. Among those were genes encoding a wide repertoire of carbohydrate-active enzymes (CAZymes) including 33 glycoside hydrolases (GH) and 87 glycosyltransferases (GT) affiliated with 17 and 12 CAZy families, respectively. DNA G+C content was 65.6 mol%. PX52T displayed only 86.0–89.8 % 16S rRNA gene sequence similarity to taxonomically described Gemmataceae planctomycetes and differed from them by a number of phenotypic characteristics and by fatty acid composition. We, therefore, propose to classify it as representing a novel genus and species, Limnoglobus roseus gen. nov., sp. nov. The type strain is strain PX52T (=KCTC 72397T=VKM B-3275T).

    Abstract. Oxygen optode measurements on floats and gliders suffer from a slow time response and sensors that are or eventually run out of calibration. Based on two dual-O2 Argo floats, we show how to post-correct for the effect of the optode’s time response and give an update on optode in-situ drift stability and in-air calibration. Both floats are equipped with an unpumped Aanderaa 4330 optode and a pumped Sea-Bird SBE63 optode. Response times for the pumped SBE63 were derived after Bittig et al. (2014). Also their methods were used to correct the time response bias. Using both optodes on each float, the time response regime of the unpumped Aanderaa optode was characterized more accurately than previously possible. Response times for the pumped SBE63 are in the range of 25–40 s, while they are between 60–95 s for the unpumped 4330 optode. Our parameterization can be employed to post-correct the slow optode time response on floats and gliders. After correction, both sensors agree to within 2–3 μmol kg−1 (median difference) in the strongest gradients (120 μmol kg−1 change over 8 minutes or 20 dbar) and better elsewhere. However, time response correction is only possible if measurement times are known, i.e., provided by the platform as well as transmitted and stored with the data. The O2 in-air measurements show a significant in-situ optode drift of −0.4 % yr−1 and −0.2 % yr−1, respectively. Optode in-air measurements are systematically biased high during mid-day surfacings compared to dusk, dawn, and nighttime. While preference can be given to nighttime surfacings to avoid this in-air calibration bias, we suggest a parameterization of the daytime effect as function of the sun’s elevation to be able to use all data and to better constrain the result. Taking all effects into account, calibration factors have an uncertainty of 0.1 %. In addition, in-air calibration factors vary by 0.1–0.2 % when using different reanalysis models as reference. The overall accuracy that can be achieved following the proposed correction routines is <1 μmol kg−1.

    ABSTRACT The cell shape of Trypanosoma brucei is influenced by flagellum-to-cell-body attachment through a specialised structure – the flagellum attachment zone (FAZ). T. brucei exhibits numerous morphological forms during its life cycle and, at each stage, the FAZ length varies. We have analysed FLAM3, a large protein that localises to the FAZ region within the old and new flagellum. Ablation of FLAM3 expression causes a reduction in FAZ length; however, this has remarkably different consequences in the tsetse procyclic form versus the mammalian bloodstream form. In procyclic form cells FLAM3 RNAi results in the transition to an epimastigote-like shape, whereas in bloodstream form cells a severe cytokinesis defect associated with flagellum detachment is observed. Moreover, we demonstrate that the amount of FLAM3 and its localisation is dependent on ClpGM6 expression and vice versa. This evidence demonstrates that FAZ is a key regulator of trypanosome shape, with experimental perturbations being life cycle form dependent. An evolutionary cell biology explanation suggests that these differences are a reflection of the division process, the cytoskeleton and intrinsic structural plasticity of particular life cycle forms. Summary: Trypanosoma brucei FLAM3 is a flagellar FAZ protein. Its depletion leads to a reduction in FAZ length, which has different consequences depending on the life cycle stage of the parasite.
